Project description:Lack of cytoplasmic beta-actin gene (Actb) leads to early embryonic lethality in mice, however targeted editing of Actb coding sequence by introducing five point mutations to encode cytoplasmic gamma-actin protein does not affect mouse viability. These mice with beta to gamma actin replacement (Actbc-g mice) develop normally and show no detectable phenotypes at young age, despite complete lack of beta-actin protein. Here we investigated the effect of the replacement of beta-actin with gamma-actin in the retina.

Project description:Transcriptional profiling of human lymphoblastoid TK6 cells comparing mock irradiated cells with cells exposed 24 hours previously to 1.67 Gy HZE (1 GeV/amu iron ions accelerated at the NASA Space Research Laboratory (NSRL) of Brookhaven National Laboratory) or 2.5 Gy 137Cs gamma rays. TK6 cells were mock irradiated or exposed to HZE or gamma-rays, and RNA was harvested 24 hours later. 3 biological replicates were independently grown and harvested during three different runs at the NSRL. One replicate per array.

Little is known about relationship between transcript synthesis, abundance and decay in macrophages during the first hours after interferon gamma treatment and how these factors influence the antiviral cellular phenotype. Microarrays were used to analyse alterations in transcript synthesis and abundance in BMDM treated with Interferon Gamma for 8h.

Global gene expression in response to SARS mouse adapted variants MA-15, MA-15epsilon, and MA-15-gamma in young and aged mice

Project description:Purpose of this experiment was to study the host-transcriptional response to MA-15, MA-15epsilon, and MA-15-gamma in both young and aged mice to further understand the differences between response within each age group, and the age-related differences in response to each virus. Young (8 weeks old) and aged (1 year old) female BALB/c mice were intranasally infected with 10^5 PFU of either MA15, MA15 gamma, MA15 epsilon, or phosphate buffer solution (PBS; mock-infection). Lungs from aged mice were harvested at 12, 24, and 48 hours post-infection. Lungs from young mice were harvested at 12, 24, 48, and 96 hours post-infection. For the aged mice, 3 biological replicates were collected for microarray analysis at each time point from the infected groups. 3 mocks at 12h, 2 mocks each at 24 and 48h time points.For the young mice, 4 biological replicates were collected for microarray analysis at each time point from the infected groups.

Retinoid X receptor gamma signaling accelerates CNS remyelination

Project description:The molecular basis of CNS myelin regeneration (remyelination) is poorly understood. Here we generate a comprehensive transcriptional profile of the separate stages of spontaneous remyelination following focal demyelination in the rat CNS. White matter tracts in the rat caudal cerebellar peduncles were focally demyelinated using 0.1% ethidium bromide, the lesions were isolated using laser capture microdissection at 5, 14 and 28 days postlesion, followed by RNA extraction and Illumina beadarray analysis of differentially expressed transcripts. We found transcripts encoding retinoid acid receptor RXR-gamma is highly differentially expressed during remyelination, and that oligodendrocyte lineage cells express RXR-gamma in rat tissues undergoing remyelination and in active and remyelinated MS lesions. RXR-gamma knockdown by RNA interference or RXR-specific antagonists severely inhibit oligodendrocyte differentiation in culture. In RXR-gamma deficient mice, adult oligodendrocyte precursor cells efficiently repopulate lesions following demyelination, but display delayed differentiation into mature oligodendrocytes. Administration of the RXR agonist 9-cis-retinoic acid to demyelinated cerebellar slice cultures and to aged rats following demyelination results in more remyelinated axons. RXR-gamma is therefore a positive regulator of endogenous oligodendrocyte precursor cell differentiation and remyelination, and may be a pharmacological target for CNS regenerative therapy. 9 Samples analysed, 3 different time points each with 3 biological replicates.
