Project description:In the present study a 44k oligonucleotide salmonid microarray, qPCR and histology were used to investigate transcriptomic responses in the distal intestine during the first week of oral exposure to soybean meal in order to gain insight into early molecular response mechanisms. Intestinal tissue samples were taken on day one, two, three, five and seven after introduction of a soybean-containing diet and compared to a control group fed fishmeal. Day 1, day 2, day 3, day 5 and day 7 (n=10 per condition) post-soybean meal feeding Atlantic salmon hybridized to common reference and compared against control (day 0 of SBM feeding). Project description:The objective of the current study was to examine the effects of yeasts on intestinal health and transcriptomic profile of Atlantic salmon fed SBM-based diets in seawater. Cyberlindnera jadinii (CJ) and Wickerhamomyces anomalus (WA) yeasts were produced in-house and processed by direct heat-inactivation with spray-drying (ICJ and IWA) or autolyzed at 50 ºC for 16 h (ACJ and AWA), followed by spray-drying. Six diets were formulated, one based on fishmeal (FM), a challenging diet with 30% soybean meal (SBM) and four other diets containing 30% SBM and 10% of each of the four yeast fractions (i.e., ICJ, ACJ, IWA and AWA). The results showed that the inclusion of CJ yeasts reduced loss of supranuclear vacuolization, along with reduction in population of CD8α positive cells present in the lamina propria of fish fed the SBM diet. The CJ yeasts controlled the inflammatory profile of fish fed SBM through up-regulation of pathways related to wound healing and taurine metabolism. Additionally, the WA yeasts dampened the inflammatory profile of fish fed SBM through down-regulation of pathways related to toll-like receptor signaling, C-lectin receptor, cytokine receptor and signal transduction. The results suggest that yeasts could be used as protein ingredients with functional properties in diets for Atlantic salmon.

Project description:The processing of seafood for human or animal consumption creates vast amounts of by-product, which is often considered waste. Although some of these by-products are used as fishmeal, bone-meal or fertilizer, as a whole, it remains under utilized. Significant amounts of proteins, lipid fractions, vitamins, and other bioactive molecules are present in these by-products, all with potential beneficial properties that could be used as alternatives to fishmeal or as supplements for aquaculture species. In an attempt to investigate their potential benefit in Atlantic salmon fish nutrition, nine experimental diets were formulated using by-products originating from various seafood processing plants. A control basal diet (no by-product added) was also formulated. Juvenile Atlantic salmons were fed one of the nine experimental diets (30% marine by-product, 70% basal diet) or the basal diet and hepatic gene expression profiling was done on fish fed each diet after 14 and 56 days. Analysis of hepatic gene expression revealed a significant amount of differentially expressed genes for each diet with roles in various pathways and biological processes. By comparing differences in hepatic gene expression levels with the nutritional composition of the various feeds, we were able to identify a number of nutritional elements that affect specific gene families. This information will be very useful for the formulation of novel fish feeds, which may be designed with specific aims, such as rapid growth, increased immunity or better general health

Project description:After a 14-week feeding trial, Atlantic salmon fed a diet based on animal by-product meal and vegetable oil (ABP) showed lower growth rates compared to others fed an FM/FO-based diet (MAR). Interestingly, feeding a vegetable meal/oil-based diet (VEG) resulted in growth rates similar to the FM/FO-based diet. A 44K microarray experiment conducted on liver total RNA samples from these three dietary groups identified 46 genes related to metabolic and growth-relevant pathways, 25 immune-related, and 12 related to oxidation-reduction processes.

Project description:Fish were fed a standard fish meal (FM) diet or a diet with partial replacement of FM with solvent extracted camelina meal (CM) (8%, 16% or 24% CM inclusion) during a 16-week feeding trial. A significant decrease in growth performance was seen in fish fed the CM inclusion diets. A 44k oligonucleotide array experiment was used to identify any differentially expressed transcripts in the distal intestine of the fish fed the 24% CM diet compared to the control. The expression level of these genes was validated using quantitative polymerase chain reaction, which was also used to measure transcript expression in the fish fed the 8% CM and 16% CM diets. Histopathological analysis was used to quantify any physical signs in inflammation in the distal intestine of the Atlantic salmon fed the CM-containing diets.

Project description:The aim of the present study was to generate an experimental model to characterize the nutrigenomic profile of a plant-derived nutritional stress (S30 = 300 g Kg-1 Soybean Meal). Our results provided: a) a snapshot of molecular signatures describing a chronic and advanced nutritional stress to which future nutrigenomic studies might refer to; and b) a platform for the identification of candidate genes for the molecular phenotyping of several physiological parameters in liver and distal intestine. Atlantic salmon was used as a model. The nutritional stress was induced by inclusions of dietary defatted soybean meal (SBM) up to a level of 300 g kg-1, being this ingredient extensively demonstrated to induce reduced performance and enteropathy in the distal intestine (Baeverfjord and Krogdahl, 1996;Urán et al., 2009;URÁN et al., 2008). A control treatment with no SBM (S0) as well as intermediate levels of inclusion (100 g kg-1 and 200 g kg-1 SBM) were included to span a range of optimal and sub-optimal conditions. Performance parameters were measured and impaired growth was taken as an indicator of pronounced and chronic nutritional stress. Molecular analyses were performed in two tissues, liver and distal intestine. Distal intestine was chosen for being the site most morphologically and physiologically affected during the development of intestinal pathologies associated with plant ingredients such as SBM (Baeverfjord and Krogdahl, 1996;Kortner et al., 2011), while liver for being arguably the most metabolic active tissue. These tissues have been the most investigated targets in nutritional studies on fish so far. To the best of our knowledge, this study is the most comprehensive of its kind to report on the transcriptomic profile of the distal intestine and the liver, hence highlighting the supporting role of this tissue, in fish undergoing SBM-induced nutritional stress. Skugor et al (, 2011) described gene expression profiles of liver and intestine in fish fed 200 g kg-1 SBM inclusion using a 11K trout array. By investigating a larger number of probes (44K) in a more severe nutritional stress (300 g kg-1), our work will add further nutrigenomic information to the current literature.

Project description:This study was performed to investigate assess the impacts of CO and/or CM containing diets on Atlantic salmon hepatic gene expression in order to identify candidate molecular biomarkers of responses to camelina-containing diets. Atlantic salmon were fed diets with complete or partial replacement of FO and/or FM with camelina oil (CO) and/or camelina meal (CM) in a 16-week trial (Control diet: FO; Test diet: 100% FO replacement with CO, with solvent-extracted FM and inclusion of 10% CM (100COSEFM10CM). A 44K microarray experiment identified liver transcripts that responded to 100COSEFM10CM (associated with reduced growth) compared to FO controls at week 16.

Project description:Piscirickettsiosis or salmonid rickettsial septicemia (SRS) caused by intracellular Gram-negative bacterial pathogen, Piscirickettsia salmonis, constitutes one of the main infectious diseases in salmonid aquaculture. In the present study, we aimed to explore the transcriptomic responses in the Atlantic salmon kidney following a low dose of EM-90-like P. salmonis infection using the consortium for Genomic Research on All Salmonids Project (cGRASP)-designed Agilent 44K salmonid oligonucleotide microarray. Two infection level phenotypes [low (L-SRS) and high (H-SRS) infection level] were revealed in infected individuals at 21 days post-injection (DPI) based on multivariate analyses of expression of four antibacterial biomarker transcripts (campb, hampa, il8a, tlr5a) and pathogen level measured by qPCR. Five fish from each group (Control, L-SRS, and H-SRS) were selected for transcriptome profiling. We identified 1636 and 3076 differentially expressed probes (DEPs) in the L-SRS and H-SRS groups, respectively (FDR = 0.01).

Project description:Black soldier fly larvae meal (BSFL) from Hermetia illucens is a promising alternative protein source in diets for farmed fish. The larvae can efficiently convert low-value organic material into high quality protein in a production cycle with low arable land and freshwater inputs. A few recent studies have shown that BSFL is a suitable protein source for Atlantic salmon (Salmo salar) in smaller controlled experiments. However, industry-relevant field trials conducted under large scale near-commercial conditions over a longer period are lacking. In this study, a feeding trial was performed to evaluate the impact of BSFL on growth performance and health of Atlantic salmon during the grow out phase in seawater, in a commercial site in Vestland county, Norway. A total of 320,000 post-smolt Atlantic salmon were distributed into six duplicate sea cages and fed one of three diets (commercial-like control diet and two test diets partially replacing the protein content of the control diet with 4 % and 8 % defatted BSFL meal) for 21 weeks, until a relevant commercial slaughter size of 4.5-5.0 kg was reached. Health parameters were assessed including histology of the distal intestine (DI), in addition to DI microbiota identification (by 16s rRNA-seq) and salmon RNA-seq of DI and head kidney (HK). The results showed that the inclusion of BSFL meal supported growth performance and had no adverse effect on gut health. The beta diversity of the distal intestine microbiota and the relative abundance of families Lactobacillaceae and the chitinolytic Bacillaceae increased in the fish fed the BSFL diets. Additionally, no histopathological changes were attributable to BSFL meal intake. Results from RNA-seq in DI revealed that BSFL inclusion modulates metabolic processes associated with lipids, the response to estrogens, the activity of immune receptors (to chemokines), phagocytosis and extracellular vesicles. Based on these results, black soldier fly larvae meal is a suitable alternative protein ingredient in inclusions of up to at least 8 % for Atlantic salmon under industrial fish farming conditions.
