Project description:Excessive accumulation of lipids in the adipose tissue is a major problem in the present-day broiler industry. However, few studies have analyzed the expression of adipose tissue genes that are involved in pathways and mechanisms leading to adiposity in chickens. Gene expression profiling of chicken adipose tissue could provide key information about the ontogenesis of fatness and clarify the molecular mechanisms underlying obesity. Chicken Genome Arrays were used to construct an adipose tissue gene expression profile of 7-week-old broilers, and to screen adipose tissue genes that are differentially expressed in lean and fat lines divergently selected over eight generations for high and low abdominal fat weight. Project description:The aim of this study was to identify genes involved in the variation of the muscle glycogen content at death (estimated through the glycolytic potential, GP), a determining factor of meat quality in chicken. Gene expression profiles of Pectoralis major muscle were established using microarrays. We compared Fat and Lean chickens issued from two lines divergently selected for abdominal fatness and also differed for muscle GP. A total of 197 genes were differentially expressed between Fat and Lean pure chickens. Several of these genes were validated by qRT-PCR. For the genes with human orthologs, annotation analyses were performed and mainly revealed pathways involved carbohydrate, fatty-acid, and protein metabolism. The relationship between gene expression and meat quality has to now be validated by further e-QTL studies on the F2 population. 8 samples from Fat chickens were compared to 8 samples from Lean chickens, 4 of these were dye-swapped.

Project description:Domestic chicken has been intensively studied because of its role as an efficient source of lean meat. However, commercial broilers resulting from genetic selection for rapid growth demonstrate detrimental traits, such as excess deposition of abdominal adipose tissue, metabolic disorders, and reduced reproduction. Therefore fast-growing broilers represent “obese” chickens compared to slow-growing egg layers (e.g, Leghorn) or wild strain of meat-type chickens (e.g., Fayoumi). Fayoumi chickens, originating from Egypt, represent a harder stain of chickens, which are more resistant to diseases. Leghorn chickens are the original breed of commercial U.S layers. Both lines were maintained highly inbred by Iowa State University poultry geneticists with an inbreeding coefficient higher than 0.95. Both Fayoumi and Leghorn demonstrated lean phenotype compared to broilers, and these three lines of chickens are genetically distant from each other. In this study, we used affymetrix microarray to profile global gene expression of three distinct genetic lines of chickens to identify functional pathways associated with leanness of domestic chickens.

Experiment Overall Design: Birds were slaughtered at 7 weeks and abdominal fat was isolated, immediately frozen in liquid nitrogen and stored at -80oC. The ten birds used in the present study were chosen by the percentage of abdominal fat (AFP): five had the highest AFP and the other five had the lowest for RNA extraction and hybridization on Affymetrix microarrays..

Project description:Obesity is a growing health concern in the United States and has been linked to a global epidemic--the metabolic syndrome. The purpose of this project was to use microarray analysis to unravel the genetic circuits controlling deposition and metabolism of fat in a hormonally-induced obesity model. Glucose, triglyceride, and free fatty acid levels of four-week-old chickens were dramatically altered after acute infusion (six days) of exogenous corticosterone (CS), effectively producing a fat phenotype. A lean phenotype was induced by thyroid hormone (T3), and the interaction of both hormones (CS+T3) was also examined. The Del-Mar 14K Chicken Integrated Systems Microarray (Geo Platform GPL1731) was used to identify differentially expressed hepatic genes (false discovery rate, P < 0.05). In the contrast of fat (CS) and lean (T3) phenotypes, 231 genes were up-regulated by CS, whereas 532 genes were up-regulated by T3. This study revealed several transport proteins, transcription factors, and metabolic enzymes that control lipogenic (CS induced) and lipolytic (T3 induced) pathways. Also, the divergent expression of three genes belonging to the β-defensin family (DEFB9, DEFB10, and DEFB11) suggests a novel role for these antimicrobial peptides in adiposity. Project description:The objective of this study was to decipher the molecular basis of feed efficiency in meat-type chicken using duodenum tissues from a chicken population divergently selected for residual feed intake (RFI). Residual feed intake is the deviation of expected feed intake from actual feed intake. Chickens that consume less feed than expected are efficient (LRFI) and chickens that consume more feed than expected are inefficient (HRFI). A divergent selection for RFI was undertaken using an unselected random bred chicken population. RFI at day 35-42 was used as a criterion for selecting low (LRFI) and high (HRFI) RFI. Duodenum tissues were collected from 16 male chickens under sterile conditions experimentation. Tissues were collected from 4 males at days 35 and 42 in each line.

Project description:Hypothalami from Fat (FL) and Lean lines (LL) of juvenile broiler chickens were obtained by divergent selection for high or low levels of abdominal fat at SRA-INRA, France. Gene expression patterns were measured during the development of adiposity at 1 to 7 weeks of age. Differentially expressed genes associated with line, age, or line-by-age were identified. Various phenotypic and metabolic alterations are present between the lines (i.e., abdominal fat, T3 levels and glycemia), however there are no alterations in ad libitum food intake between the lines. A balanced block hybrdization design and the Del-Mar 14K chicken integrated systems microarrays were used to measure gene expression during development. 561 genes were differentially expressed between line, and 442 genes were significant for line-by-age interactions. The greatest number of genes were differentially expressed at week 1 of age, prior to any divergence in adiposity between the two lines.
