Mouse spleens were harvested and cells FACS sorted into various immune cells containing bacteria. Short reads were mapped to the S. enterica 14028 genome. Mice were infected with S. enterica strain 14028. Spleens were collected, homogenized and cells FACS sorted. Total RNA was extracted using the Ambion mirVana RNA isolation kit. Purified RNA was depleted of rRNA utilizing the epicentre ribozero metabacteria and eukaryotic kits.

Subcellular proteomic survey of Salmonella

Project description:The project performed a subcellular proteomic analysis of Salmonella enterica subsp. enterica serovar Typhimurium str. 14028 grown under standard laboratory and phagosome-mimicking conditions in vitro. Analysis of proteins from cytoplasmic, inner membrane, periplasmic, and outer membrane fractions yielded coverage of 25% of the theoretical proteome. Confident subcellular location could be assigned to over 1000 proteins, with good agreement between experimentally observed location and predicted/known protein properties. Comparison of protein location under the different environmental conditions provided insight into dynamic protein localization and possible moonlighting (multiple function) activities.

Single-cell profiling identifies ACE+ granuloma macrophages as a non-permissive niche for intracellular bacteria during persistent Salmonella infection

Project description:Macrophages mediate key antimicrobial responses against intracellular bacterial pathogens, such as Salmonella enterica. Yet, they can also act as a permissive niche for these pathogens to persist in infected tissues within granulomas, which are immunological structures comprised of macrophages and other immune cells. We apply single-cell transcriptomics to investigate macrophage functional diversity during persistent Salmonella enterica serovar Typhimurium (STm) infection in mice. We identify determinants of macrophage heterogeneity in infected spleens and describe populations of distinct phenotypes, functional programming, and spatial localization. Using a STm mutant with impaired ability to polarize macrophage phenotypes, we find that angiotensin converting enzyme (ACE) defines a granuloma macrophage population that is non-permissive for intracellular bacteria and their abundance anticorrelates with tissue bacterial burden. Disruption of pathogen control by neutralizing TNF is linked to preferential depletion of ACE+ macrophages in infected tissues. Thus ACE+ macrophages have limited capacity to serve as cellular niche for intracellular bacteria to establish persistent infection.

Salmonella Typhimurium ATCC14028 fimZ mutant

Project description:Fimbriae are hair-like structures present on the outer membrane of many Enterobacteriaceae and such appendages are proposed to play a significant role in the attachment of the bacteria to host cells and tissues. Salmonella enterica serovar Typhimurium has the potential to produce 13 different fimbrial types, among which type 1 fimbriae is the most common found. Expression of type 1 fimbriae is cooperatively controlled by fimZ, fimY, stm0551, fimW, and fimU within the fim gene cluster. FimZ belongs to the response regulator of the two-component regulatory system in bacteria and functions as a DNA binding protein. FimZ is a positive regulator for type 1 fimbrial expression in S. Typhimurium. A fimZ mutant in ATCC 14028 strain constructed by allelic exchange did not produce type 1 fimbriae. Total RNA was extracted from the parental ATCC 14028 and its fimZ mutant cultured in static broth and analyzed by hybridization to a S. Typhimurium LT2 DNA microarray. Transcriptomic analysis indicated that the type 1 fimbriae related genes, multidrug efflux protein gene acrF were down-regulated, whereas flagella associated genes, chemotaxis and virulence genes such as invF and invH genes were up-regulated in the fimZ mutant strain. It is possible that FimZ protein may play a role to interact with other genes besides regulating type 1 fimbriae.

Dendritic cells infected with Salmonella enterica

Project description:Salmonella enterica causes serious global burden of morbidity and mortality and is a major cause of infant bacteremia in sub Saharan Africa. Diseases caused by Salmonella are treatable with antibiotics but successful antibiotic treatment has become difficult due to antimicrobial resistance. An effective vaccine together with public health effort may therefore be a better strategy to control these infections. Protective immunity against Salmonella depends primarily on T cell-mediated immune responses and therefore identifying relevant T cell antigens is necessary for Salmonella vaccine development. Our laboratory has used an immunoproteomics approach to identify Chlamydia T cell antigens that exhibited significant protection against Chlamydia infection in mice. In this study, we infected murine bone marrow derived dendritic cells from C57BL/6 mice with Salmonella enterica strain SL1344 followed by isolation of MHC class I and II- molecules and elution of bound peptides. The sequences of the peptides were then identified using tandem mass spectrometry. We identified 87 MHC class II and 23 MHC class I Salmonella derived peptides. Four of 12 peptides stimulated IFN-? production by CD4 T cells from the spleens of mice with persistent Salmonella infection. These antigens will be useful for Salmonella immunobiology research and are potential Salmonella vaccine candidates.
