Transcriptome sequencing analysis of swamp eel

ABSTRACT: The swamp eel or rice field eel (Monopterus albus) taxonomically belongs to the family Synbranchidae of the order Synbranchiformes (Neoteleostei, Teleostei, Vertebrata). It is not only an economically important freshwater fish in aquacultural production, but also an increasingly known model species for biological studies. Understanding molecular mechanisms underlying sex change is a major area of interest. The swamp eel thus offers a powerful system for studying sexual development and adaptive evolution in vertebrates.The whole genome sequencing provides valuable resources for sex control in fish production, species protection through manipulating sex reversal genes, and potentially enabling effective population control and promoting reproduction health in human.

Project description:Vibriosis caused by Vibrio vulnificus on eels represents an important threat for this specie under culture conditions. Development of new transcriptomic tools is essential to increase the knowledge of eel biology, that nowadays is scarcer. Therefore, using previous results obtained by 454 sequencing of the eel immune-enriched transcriptome, an eel-specific custom microarray have been designed. Gills transcriptomic pattern were analyzed as a principal portal of entry for pathogens in fish after 1h of bath infection with Vibrio vulnificus to describe gill immune response. Moreover, two different strains were used, vibro vulnificus wild type (R99) and rtx double mutant (CT285), to asses the virulence of these pathogen caused by MARTX. Adult european eels were bath infected with two Vibrio vulnificus strains, the wild type and double Rtx mutant (CT285). After 0, 3, 12h post-infection eel gills were sampled. Three individuals per experimental point were sampled, including a Control group and a Handling control group. Obtaining a total of 24 samples. The transcriptomic profile was described for each individual sample.

Blood plasma proteomic profiles of pink salmon Oncorhynchus gorbuscha Walbaum, 1792 during spawning migration

Project description:The pink salmon (Oncorhynchus gorbuscha) is a commercial anadromous fish species of the family Salmonidae. The species has a peculiar life cycle that includes spawning migration from marine to freshwater environments, which is accompanied by significant adaptive changes in the body, both the physiological and biochemical. This study described and revealed the variability of blood plasma proteomes of female and male pink salmon collected from three different biotopes - marine, estuarine and riverine - that the fish pass through spawning migration. Identification and comparative analysis of pink salmon blood plasma protein profiles were performed using proteomic and bioinformatic approaches. Blood proteomes of female and male spawners collected from different biotopes were qualitatively and quantitatively distinguished. Females differed primarily by proteins associated with reproductive system development (certain vitellogenin and choriogenin), lipid transport (fatty acid binding protein) and energy production (fructose 1,6-bisphosphatase), and males - by proteins involved in blood coagulation (fibrinogen), immune response (lectins) and reproductive processes (vitellogenin). Differentially expressed sex-specific proteins were implicated in proteolysis (aminopeptidases), platelet activation (β- and γ-chain fibrinogen), cell growth and differentiation (a protein containing the TGF_BETA_2 domain) and lipid transport processes (vitellogenin and apolipoprotein). The results obtained are of fundamental and practical importance, providing to the existing knowledge of biochemical adaptations to spawning of pink salmon, representative of the economically important migratory fish species.

Project description:Sequential hermaphroditism, whereby the sex of one individual changes during its development, has been described in many fish species. Yet, the genetic regulation of this dynamic process of sexual differentiation is not well understood. Here, we report the characterization of sequential hermaphroditism in an aquacultural fish, the zig-zag eel (Mastacembelus armatus), based on 4 years of artificial breeding. All M. armatus individuals initially developed as females, with approximately half of them then becoming males after going through an intersex phase.

Proteomic studies on the swimbladder of the European eel (Anguilla anguilla)

Project description:The swimbladder of fish is a vital organ that along with gas glands in the swimbladder wall enables key physiological functions including buoyance, diving and coping with different hydrostatic pressures. Specific intramural glands provide gas for regulating gas pressure in the swimming bladder. Transcriptomic analysis have revealed evidence at the RNA level but no specific studies at the protein level have been carried out so far. Herein, it was the aim of the study to show swimbladder proteins of the European eel by label-free LC/MS (Q-Exactive Plus) that resulted in the identification of 6,223 protein groups. Neurotransmitter receptors and -transporters were enriched in the membrane fraction and enzymes for gas production were observed. The list of identified proteins may represent a useful tool for further proteomics experiments on this organ.

Transcription profiling of brain from freshwater and sea water adapted silver eels, Anguilla anguilla

Project description:We investigated salinity adaptation during the migration from freshwater to seawater of European eel (Anguilla anguilla) by examining the hypothesis that: The brain is the central organ for the co-ordination of environmental cues (day length, photoperiod, temperature and environmental salinity) with the anatomical and physiological adaptations which accompany pre-migrational morphogenesis and the osmoregulatory plasticity seen in post-migrational, salinity-adapted fish. We have characertised the mRNA expression profiles for the brains of fresh water and sea water adapted silver eel using a highly representative brain cDNA microarray. The array comprises 5760 cDNA clones from A.anguilla ranging from 0.5 -10 kb and an estimated redundancy of > 5 %.

Changes in ovarian gene expression profiles in maturing European eel (Anguilla anguilla)

Project description:The goal of this study was to gain a better understanding of the genetic background of gonadal maturation of the European eel and to use gene expression profiles to identify predictive markers for broodstock selection that can be measured in blood samples. To find leads for maturation markers we performed a pilot deep-sequencing transcriptome analysis of ovarian tissue derived from a yellow eel, a prepubertal silver eel and a post-spawning matured eel. Among the best leads were two key players in steroidogenesis, namely pP450c17 and liver receptor homolog-1.
