Project description:Live fish transport often results in mass mortality, but the underlying mechanisms are not well known. Largemouth bass (LMB, Micropterus salmoides) juveniles were subjected to 4-hour transportation followed by 5 days of recovery. Mortality (33.61 ± 10.58 %) of the transported fish mainly occurred between 2 and 3 days of recovery. Except for the dead fish, the survivors demonstrating abnormal symptoms were considered more sensitive to the transport stress than the normal ones. Liver of the abnormal fish had significantly (p < 0.05) lower glutathione (GSH) content and superoxide dismutase (SOD) activity, and irregular cellular morphology. The liver and spleen collected before transportation (T0), at the end of transportation (Te), after 1 day of recovery (R1d), and after 3, 4 and 5 days of recovery (R3d, R4d and R5d, from both the normal and abnormal fish) were subjected to RNA-sequencing. Time series gene expression data of the T0, Te, R1d, R3d-R5d (from the normal fish) samples were analyzed to explore transcriptional responses elicited by the transport stress. Transportation-activated transcriptional regulations could be classified as immediate response (Te), and early (R1d) and late recovery (R3d to R5d) phase in both tissues. High tissue specificity was found for functional enrichments of the up-regulated genes. The up-regulated genes of the liver are highly enriched in cholesterol, sterol and steroid biosynthesis, while those of the spleen are primarily associated with hemopoiesis and immune response. However, large numbers of functional enrichments were shared by the down-regulated genes of the liver and spleen, especially during the recovery phase. Representatives of the depressed processes included neurogenesis, angiogenesis and cell migration, etc. Moreover, large numbers of differentially expressed genes (DEGs) were identified between the normal and abnormal fish. Functional enrichment analyses for the DEGs revealed that failing to terminate the stress responses in the spleen and over depression of the physiological roles of the liver are critical for stress susceptibility of the abnormal fish. Furthermore, an array of core genes associated with stress susceptibility were identified. Our data provide novel insights into the genetic factors underlying susceptibility of LMB to the transport stress.

Project description:Transport stress is a big threat to most teleost fish during production practice, causing mass loss to the aquaculture industry. Fish gills are mucosa-associated lymphoid tissue that directly contacted with water, which is the good choice to study the mechanism of transport stress. Gills transcriptome results revealed 1551 differentially expressed genes (744 up-regulated and 807 down- regulated) between the control and 16h transportation groups. The top 10 enriched KEGG pathways are all immune-related. After qRT-PCR validation, we found that the toll-like receptors and nod-like receptors signaling pathways mediate the gill’s immune response to transport stress: tlr5, tnfɑ, hsp90ɑ, il-1ß and map2k4 were significantly up-regulated and arrdc2 was significantly down-regulated under transport stress. These results suggested that the transport stress altered innate immunity responses and induced metabolic changes in gills of hybrid yellow catfish. In conclusion, the results provide new perspectives on immune response in yellow catfish against transport stress and provide the possibility of mining resistance genes for future optimize transportation plan.

Project description:The transport of live fish is a routine operation in aquaculture. However, the effects of transport stress on intestines gene expression profile is poorly understood. The results from the transcriptome analysis revealed that most differentially expression genes (DEGs) were enriched in immune and metabolic related signaling pathways. Toll like receptors is a potential pathway in immune response induced by transport stress. Changes in genes related to lipid metabolism may be related to the increased metabolic expenditure for the normal body functions in a response to transport stress. In addition, changes in gene expression in the control, transport, and recovery groups were revealed that transport-induced local immune damage is reversible while metabolic disorder has hysteresis. These results provide new perspectives on immune and metabolic response in yellow catfish against transport stress and theoretical support for future optimization of their transportation.

Project description:Hepatic metabolic adjustments are key adaptive mechanisms to stress in fish targeting at increasing energy availability for the animal to efficiently cope with a stressor. A comparative gel-based proteomics analysis (2D-DIGE) was employed to discover the set of liver proteins involved in the adaptive processes that tune the physiological response to chronic stressors in Sparus aurata. Three separated trials were established where fish were submitted to different stressors (overcrowding, net handling and hypoxia). Two intensities for each stressor were tested, in triplicate tanks. Two liver samples per tank (6 per experimental treatment) were collected for liver proteome analysis at the end of each trial.

Project description:Circulating corticosteroids orchestrate stress adaptation, including inhibition of inflammation. While pathways governing corticosteroid biosynthesis and intracellular signaling are understood, less is known about mechanisms controlling plasma corticosteroid transport. Here, we show hepatocyte KLF15 (Kruppel-like factor 15) controls plasma corticosteroid transport and inflammatory responses through direct transcriptional activation of Serpina6, which encodes corticosteroid binding globulin (CBG). Klf15-deficient mice have profoundly low CBG, reduced plasma corticosteroid binding capacity, and heightened mortality during inflammatory stress. These defects are completely rescued by reconstituting CBG, supporting that KLF15 works primarily through CBG to control plasma corticosterone homeostasis. To understand transcriptional mechanisms, we generated the first KLF15 cistromes using newly engineered Klf153xFLAG mice. Unexpectedly, liver KLF15 is predominantly promoter-enriched, including Serpina6, where it binds a palindromic GC-rich motif, opens chromatin, and transactivates genes with minimal associated gene repression. Overall, we provide new mechanistic insight into KLF15 function and identify a hepatocyte-intrinsic transcriptional module that potently regulates systemic corticosteroid transport and inflammation.

Project description:High-throughput proteomics was used to determine the role of the fish liver in defense responses to bacterial infection, done using a rainbow trout (Oncorhynchus mykiss) model following infection with Aeromonas salmonicida, the causative agent of furunculosis. The vertebrate liver has multifaceted roles in innate immunity, metabolism, and growth; we hypothesize this tissue serves a dual function in supporting host defense in parallel to metabolic adjustments that promote effective immune function. While past studies have reported mRNA responses to A. salmonicida in salmonids, the impact of bacterial infectionon the liver proteome remains uncharacterized in fish.

Project description:In cell models, changes in the “accessible” pool of plasma membrane (PM) cholesterol are linked with the regulation of ER sterol synthesis and metabolism by the Aster family of nonvesicular transporters. However, the relevance of such nonvesicular transport mechanisms for lipid homeostasis in vivo has not been defined. Here we reveal two physiological contexts that generate accessible PM cholesterol and engage the Aster pathway in liver: fasting and reverse cholesterol transport (RCT). During fasting, adipose tissue–derived fatty acids activate hepatocyte sphingomyelinase to liberate sequestered PM cholesterol. Aster-dependent cholesterol transport during fasting facilitates cholesteryl ester (CE) formation, cholesterol movement into bile, and VLDL production. During RCT, HDL delivers excess cholesterol to the hepatocyte PM through SR-BI. Loss of hepatic Asters impairs cholesterol movement into feces, raises plasma cholesterol levels, and causes cholesterol accumulation in peripheral tissues. These results reveal fundamental mechanisms by which Aster cholesterol flux contributes to hepatic and systemic lipid homeostasis.

Project description:Schizophrenia (SCZ) is a common, disabling mental illness with high heritability but complex, poorly understood genetic etiology. As the first phase of a genomic convergence analysis of SCZ, we generated 16.7 billion nucleotides of short read, shotgun sequences of cDNA from post-mortem cerebellar cortices of 14 patients and six matched controls. A rigorous analysis pipeline was developed for analysis of digital gene expression studies. Sequences aligned to approximately 33,200 transcripts in each sample, with average coverage of 450 reads per gene. Following adjustments for confounding clinical, sample and experimental sources of variation, 215 genes differed significantly in expression between cases and controls. Golgi apparatus, vesicular transport, membrane association, Zinc binding and regulation of transcription were over-represented among differentially expressed genes. Twenty three genes with altered expression and involvement in presynaptic vesicular transport, Golgi function and GABAergic neurotransmission define a unifying molecular hypothesis for dysfunction in cerebellar cortex in SCZ.
