Project description:Functional characterization of the Xap5 protein using epistatic genetic interaction analysis (E-MAP), RNA-seq and ChIP-chip along with other growth experiments. wild type and mutant cells were grown at 37C in EMM50 and RNA was extracted from exponentially growing cells. Strand specific libraries were constructed with random primers. Ribo zero was used to remove r-RNA. For ChIP-chip Agilent 60mer array was used to analyze DNA recovered by chromatin immunoprecipitation of Xap5 from fission yeast culture.

Transcription profiling of fission yeast Pht1 (H2A.Z) deletion mutants

Project description:Histone variant H2A.Z has a conserved role in genome stability, although it remains unclear how this is mediated. Here we demonstrate that the fission yeast Swr1 1 ATPase inserts H2A.Z (Pht1) into chromatin and Kat5 acetyltransferase (Mst1) acetylates it. Deletion or an unacetylatable mutation of Pht1 1 leads to genome instability, primarily caused by chromosome entanglement and breakage at anaphase. This leads to the loss of telomere-proximal markers, though telomere protection and repeat length are unaffected by the absence of Pht1. Strikingly, the chromosome entanglement in pht1? anaphase cells can be rescued by forcing chromosome condensation before anaphase onset. We show that the condensin complex, required for the maintenance of anaphase chromosome condensation, prematurely dissociates from chromatin in the absence of Pht1. This and other findings suggest an important role for H2A.Z in the architecture of anaphase chromosomes.

H2A.Z (pht1) ChIP-seq data in 968, Y0036, and swc5-del strains

Project description:After finding a QTL hotspot in a fission yeast cross of strains 968xY0036 that was apparently driven by frame shift mutation in swc5 (swc5-fs), we have determined the genome-wide occupancy of the histone variant H2AZ (PhT1). Indeed swc5 has been shown to affect H2AZ occupancy. To test the impact of swc5-fs on H2A.Z deposition, we performed genome-wide chromatin immuno-precipitation of H2A.Z coupled with deep sequencing (ChIP-seq) in the two parental strains (968, Y0036) and in a swc5 deletion strain. We also assed the histone H3 occupancy as a positive control. We generated pht1-myc tagged strains to precipitate H2AZ with Myc antibody. Several negative controls have been performed, in particular pulldowns with the non tagged strains. Results show a reduced H2A.Z occupancy at the +1 histone in both Y0036 and swc5-deletion strains.

Project description:Chromatin regulatory proteins affect diverse developmental and environmental response pathways via their influence on nuclear processes such as the regulation of gene expression. Through a genome-wide genetic screen, we implicate a novel protein called X-chromosome-associated protein 5 (Xap5) in chromatin regulation. We show that Xap5 is a chromatin-associated protein acting in a similar manner as the histone variant H2A.Z to suppress expression of antisense and repeat element transcripts throughout the fission yeast genome. Xap5 is highly conserved across eukaryotes, and a plant homolog rescues xap5 mutant yeast. We propose that Xap5 likely functions as a chromatin regulator in diverse organisms.
