Transfection was performed using lentiviral particles, RNA was harvested after culture for 2 weeks. Results provide insight into functional UGT2B17 involvement in CLL cells.
Project description:SUM52PE cells were lentivirally transduced with PKLO empty vector control (Sigma SHC001 MISSION PLKO.1-PURO CONTROL VECTOR) or shRNA specific for C11orf67 (Sigma Clone ID: NM_024684.2-280s21c1 SHCLND-NM_024684 TRCN0000263814) in biological triplicates. Differentially regulated targets by the shRNA relative to empty vector control were determined.
Project description:To study the role of SmcHD1 in regulating gene expression, we have stably knocked down SmcHD1 in HEK293 cells and looked at change in gene expression. shRNA3 and shRNA4 were used to target the expression of the SmcHD1 gene (Genbank Accession number NM_015295). The protein levels of SmcHD1 were knocked down in excess of 90% normal levels in 293 cells. shRNA NC5 a scrambled control shRNA and did not change SmcHD1 protein levels in 293 cells.
