Project description:We used laser capture microdissection to isolate different zones of the articular cartilage from proximal tibiae of 1-week old mice, and used microarray to analyze global gene expression. Bioinformatic analysis corroborated previously known signaling pathways, such as Wnt and Bmp signaling, and implicated novel pathways, such as ephrin and integrin signaling, for spatially associated articular chondrocyte differentiation and proliferation. In addition, comparison of the spatial regulation of articular and growth plate cartilage revealed unexpected similarities between the superficial zone of the articular cartilage and the hypertrophic zone of the growth plate. Project description:We used laser-capture microdissection (LCM) to isolate specific cells from the Medicago truncatula nodule meristem (M), the distal infection (DIZ), the proximal infection zone (PIZ), infected cells (IC) and uninfected cells (UIC) from the fixation zone. Based on Medicago GeneChips, we identified the cell- and tissue-specific programm of gene expression in Medicago truncatula root nodules. Nodules were harvested three weeks after inoculation of Medicago truncatula (genotype Jemalong A17) plants with Sinorhizobium meliloti strain 2011. Nodules were fixed in Farmer's fixative and subsequently embedded in paraffin. 8 M-BM-5m de-paraffined sections were used to capture cells from the nodule meristem, distal infection zone, proximal infection zone, infected cells and uninfected cells from the fixation zone, using an Arcturus Pixcell II laser capture microscope. 3 biological replicates were used for each cell-/tissue type. After RNA extraction, the RNA was amplified and used for Medicago Gene Chip hybridization.

Hippocampal dentate gyrus (DG) are more responsive than the nucleus accumbens (NAcc) following the glucocorticoid receptor overexpression in the forebrain. Laser capture microdissection was performed. Microdissected areas from the dentate gyrus or nucleus accumbens were collected from serial sections for each animal.
Project description:FTIR based spectral histopathology was used for label-free classification of tumor tissue sections. The regions of interest were identified and annotated with high spatial resolution. The spatial information was used for automated laser capture microdissection of relevant tissue areas. Subsequently, the collected tissue samples were analyzed using quantitative proteomics. This novel approach for precise sample collection in clinical studies was applied to differentiate epitheloid and sarcomatoid subtypes of diffuse malignant mesothelioma.
