MicroRNA expression signatures for developing tongue primordia in mouse embryos

ABSTRACT: We aimed to understand the molecular basis of muscular development in tongue. Project description:We aimed to understand the molecular basis of muscular development in tongue. We focused on the critical stages of tongue myogenesis, namely, E10.5 (settlement of myogenic progenitors in tongue primordium), E11.5 (initiation of myoblast differentiation with the development of lateral lingual swellings). We conducted miRNA microarray analysis of the tongue primordia at E10.5 and E11.5, during which myoblast differentiation was induced as a result of the activation of muscle regulatory gene expression. The medial part of the fused mandibular prominences that corresponds to a tongue primordium were collected from E10.5 and E11.5 mouse embryos. Forty embryos for each time-point were used to obtain a pool of total RNA.

Project description:The mouse hypoglossal nerve originates from the occipital motor nuclei and projects a long distance, reaching the vicinity of the tongue primordia, the lateral lingual swelling, at E11.5. However, the details of how the hypoglossal nerve correctly projects to the primordia are not well understood. To investigate the molecular basis of hypoglossal nerve elongation, we employed cDNA microarray technology and screened for candidate genes involved in the axon guidance event.

Expression data from mandibular arches in mouse embryos

Project description:Fusion of branchial arch derivatives is an essential event in the development of craniofacial architecture. A unique feature of the mandibular arch development is medial/lateral compartmentalization for the molecular networks. Those networks give rise to multiple region-specific organs, namely teeth, a tongue, salivary glands, and the supporting matrices such as bones and cartilages. We aimed to investigate molecular networks that govern the fusion process during mouse mandibular development. To this end, cDNA microarray technology was employed for screening of spatio-temporal gene expression in developing mandibular arch from E9.7 through E11.5. We conducted to divide a mandibular arch medially and laterally to compare both gene expression. From an embryo at E10.5, a medial (M) sample of the mandibular arch was dissected out -at just the distal end of opposed lateral lingual swellings-, and the bulk of remnant lateral region was collected as (L) sample under a stereomicroscope. Forty embryos for each time-point were used to obtain a pool of total RNA.

Genome-wide identification of Foxf2 transcriptional target genes in tongue development

Project description:The tongue is a specialized muscular organ that performs multiple essential functions including mastication, deglutition, oral sensation, oral cleansing, airway maintenance and vocalization. In this study, we show Foxf1/Foxf2 serves as key mediators of hedgehog signaling in regulating myoblast migration, differentiation, and intrinsic tongue muscle organization. We took advantage of the Foxf2FLAG mice which carries 3xFLAG epitope-tagged endogenous Foxf2 protein and characterized genome-wide Foxf2 binding sites in the developing tongues using chromatin immunoprecipitation and genome sequencing (ChIP-seq). Further analyses demonstrate that Foxf1/2 transcription factors directly control the expression of Hgf, Tgfb2, and Tgfb3, to regulate tongue myogenesis.

Genome-wide maps of methylations in experimental lingual carcinogenesis

Project description:To identification of genome wide DNA methylation profiling during lingual tumorigenesis, tongue cancer was induced by oral delivery of 4-nitroquinoline-1-oxide (4NQO) at a dose of 50mg/L for 16weeks in C57BL/6J female mice. Nine samples of lingual mucosa from 0 (encoded as C in experiment), 12 (encoded as M in experiment) and 28 (encoded as E in experiment) week, which were supposed as normality, early and advanced stage during lingual tumorigenesis, were harvested for methylated DNA Immunoprecipitation sequencing. Illumina HiSeq 2000 was used to obtain DNA methylation profiles across 16023 CpG Islands including 26600 promoters and 21625 genebodies in murine tongue samples. Samples included 3 normal lingual mucosa,3 lingual mucosa with hyperplasia, mild and moderate dysplasia, 3 lingual mucosa with carcinoma in situ and invasive cancer.

The unfolded protein response gene Ire1α is required for tissue renewal and normal differentiation in the mouse tongue and esophagus

Project description:The IRE1α-XBP1s signaling branch of the unfolded protein response is a well-characterized survival pathway that allows cells to adapt to and resolve endoplasmic reticulum stress. Recent data has broadened our understanding of IRE1α-XBP1s signaling beyond a stress response and revealed a physiological mechanism required for the differentiation and maturation of a wide variety of cell types. Here we provide evidence that the IRE1α-XBP1s signaling pathway is required for the proliferation and maturation of basal keratinocytes in the mouse tongue and esophageal epithelium. Mice with conditional targeted deletion of either IRE1α or XBP1 in keratin 14 expressing basal keratinocytes displayed severe thinning of the lingual and esophageal mucosa that rendered them unable to eat. In IRE1α null epithelium harvested at an earlier timepoint, genes regulating cell proliferation, cell-cell adhesion, and keratinization were significantly downregulated; indirect immunofluorescence revealed fewer proliferating basal keratinocytes, downregulation of E-Cadherin, and thinning of the loricrin-positive granular and cornified layers. The number of Tp63 positive basal keratinocytes was reduced in the absence of IRE1α, and expression of the Wnt pathway transcription factor LEF1, which is required for the proliferation of lingual transit amplifying cells, was also significantly downregulated at the transcript and protein level. Together these results reveal an essential role for IRE1α-XBP1s in the maintenance of the stratified squamous epithelial tissue of the tongue and esophagus.

RNA expression in tongue squamous cell carcinoma, tumour free tongue tissue and control tongue

Project description:Tongue squamous cell carcinoma is a tumour type with rather low five year survival, around 60%. The poor survival rate has been ascribed to late detection, a high frequency of locoregional recurrence, the occurrence of secondary primary tumours and death due to comorbidity. One reason for development of recurrence is thought to be the existence of transformed cells in areas adjacent to the primary tumour, cancerization field effect. The aim of this study was to map the changes in the tumour free tongue tissue adjacent to tongue tumours compared with healthy control tongue tissue to better understand the cancerization field effect. Tissue biopsies were collected from tumour (T) and tumour free tissue adjacent to the tumour (TF) from patients with tongue squamous cell carcinoma. Control tissue was collected from latter border of the tongue of tumour free healthy volunteers (C). All samples were homogenized and RNA was extracted. The RNA was biotin labelled and run on Illumina HT-12 bead chip array.

mRNA sequencing of epithelial cells from the esophagus and tongue

Project description:In this study, we used RNA sequencing to characterize a new population of taste buds found in the esophagus and compared them to their lingual counterpart. We concluded that esophageal taste buds share many markers with lingual taste buds althoug they seem to express less taste receptors in average.
