Project description:During spermiogenesis in mammals and many other vertebrate classes, histone-containing nucleosomes are replaced by protamine toroids, which can repackage chromatin at a 10 to 20-fold higher density than in a typical somatic nucleus. However, recent evidence suggests that sperm of many species, including human and mouse retain a small compartment of nucleosomal chromatin, particularly near genes important for embryogenesis. As in mammals, spermiogenesis in the fruit fly, Drosophila melanogaster has also been shown to undergo a programmed substitution of nucleosomes with protamine-like proteins. Using chromatin immunoprecipitation (ChIP) and whole-genome tiling array hybridization (ChIP-chip), supported by immunocytochemical evidence, we show that in a manner analogous to nucleosomal chromatin retention in mammalian spermatozoa, distinct domains packaged by the canonical histones H2A, H2B, H3 and H4 are present in the fly sperm nucleus. We also find evidence for the retention of nucleosomes with specific histone H3 trimethylation marks characteristic of chromatin repression (H3K9me3, H3K27me3) and active transcription (H3K36me3). Project description:Using the frog Xenopus laevis as a model system we profile epigenetic features of sperm and spermatid to study how they relate to gene expression in embryos. We observe that sperm is epigenetically programmed to regulate embryonic gene expression. By comparing the development of sperm- and spermatid-derived frog embryos we show that the programming of sperm for successful development relates to its ability to regulate transcription of a set of developmentally important genes. During spermatid maturation into sperm, these genes lose H3K4me2/3 and retain H3K27me3 marks. Experimental removal of these epigenetic marks, at fertilization, deregulates gene expression in the resulting embryos in a paternal chromatin dependent manner. This demonstrates that epigenetic instructions delivered by the sperm at fertilization are required for correct regulation of gene expression in the future embryos. The epigenetic mechanisms of developmental programming revealed here are likely to relate to the mechanisms involved in transgenerational transmission of acquired traits.

Project description:The male gamete has an immense role in reproduction. Besides contributing one half of the genomic DNA, it contributes important epigenetic information that controls and orchestrates its expression during the early stages of embryo development. Although, DNA methylation is a widely studied epigenetic modification, other epigenetic changes like histone modifications, nucleosome landscape and chromatin compaction are also gaining significant consideration. Sperm chromatin despite being majorly compacted with protamines; strategically retains 2-4% of the genome in nucleosomes. While DNA bases are resilient, epigenetic marks are highly susceptible to reprogramming. Environmental pollutants have been known to affect epigenetic marks and many of those pollutants are known to be xeno-estrogenic. Previous studies using tamoxifen, a Selective Estrogen Receptor Modulator – SERM, has been shown to be associated with post implantation loss and DNA methylation aberration in rats. Also, Cyperoterone acetate (CPA), an antiandrogen, has been shown to induce retention of histone modifications in testis. In this study, we show by MNase-sequencing that systemic hormonal disruption leads to deviation in the nucleosomal localization in spermatozoa at many genes of significance during early embryo development. Thus, environmental pollutants, which are mainly estrogenic or antiandrogenic in nature, may exert epigenetic aberrations in the spermatozoa through hormonal disruption which may lead to severe complications during embryo development or in adult life.

Project description:Here we investigated the stability of nucleosomal modifications during pull-down affinity purification with HeLa nuclear extracts. Unmodified di-nucleosomes and di-nucleosomes decorated with H3K4me3K9acK14acK18acK23acK27ac, H4K5acK8acK12acK16acK20me2 and incorporating histone variant H2A.Z were incubated with HeLa nuclear extract or buffer alone for 4 hours at 4 degrees Celsius. The relative abundances of nucleosomal modifications were quantified using LC-MS.
