Project description:The male gamete has an immense role in reproduction. Besides contributing one half of the genomic DNA, it contributes important epigenetic information that controls and orchestrates its expression during the early stages of embryo development. Although, DNA methylation is a widely studied epigenetic modification, other epigenetic changes like histone modifications, nucleosome landscape and chromatin compaction are also gaining significant consideration. Sperm chromatin despite being majorly compacted with protamines; strategically retains 2-4% of the genome in nucleosomes. While DNA bases are resilient, epigenetic marks are highly susceptible to reprogramming. Environmental pollutants have been known to affect epigenetic marks and many of those pollutants are known to be xeno-estrogenic. Previous studies using tamoxifen, a Selective Estrogen Receptor Modulator – SERM, has been shown to be associated with post implantation loss and DNA methylation aberration in rats. Also, Cyperoterone acetate (CPA), an antiandrogen, has been shown to induce retention of histone modifications in testis. In this study, we show by MNase-sequencing that systemic hormonal disruption leads to deviation in the nucleosomal localization in spermatozoa at many genes of significance during early embryo development. Thus, environmental pollutants, which are mainly estrogenic or antiandrogenic in nature, may exert epigenetic aberrations in the spermatozoa through hormonal disruption which may lead to severe complications during embryo development or in adult life.

Project description:Hormonal stimulation in common carp is a routine practice to enhance sperm production and control gamete maturation. The mechanism regulating functional changes of semen composition upon hormone treatment is unknown at present. The aim of the study was to compare proteome of carp spermatozoa and seminal plasma from control (PBS, n=10) and hormonally stimulated males (D-Ala6,Pro9-NEt)-mGnRH+metoclopramide, n=10). Comparative proteomics based on two-dimensional differential in-gel electrophoresis was employed to quantitatively analyze the spermatozoa and seminal plasma proteins changed in abundance due to hormonal stimulation. Changes in phosphorylation and oxidation level of spermatozoa proteins were also analyzed. The proteomic results were validated by western blot, enzymatic assay and mRNA expression analysis.In total we identified 62 spots (38 up and 24 down regulated) corresponding to 48 proteins with differential abundance in spermatozoa and 36 spots (23 up and 13 down regulated) corresponding to 23 proteins differentially abundant in seminal plasma upon hormone treatment (P<0.05; fold change 1.2). The majority of spermatozoa proteins changed in abundance as well as phosphorylation and carbonylation level were involved in cillium assembly and organization, energy metabolism, protein folding, and signal transduction while differentially abundant seminal plasma proteins were associated with immune and stress response, lipid metabolism and tissue remodelling. Our results strongly suggest that hormonal treatment is not only related to hydration of testis but also induces modification of spermatozoa proteins and synthesis of seminal plasma proteins. ROS signaling and cAMP/PKA and calcium/calmodulin pathways are involved in sperm response to hormonal stimulation.

Project description:The phenomenon of nucleosome retention in mammalian sperm chromatin is still not clarified yet. The goal of our study was to characterize the binding sites of sperm nucleosomes in the human and bovine genomes, and through this, to clarify whether nucleosome retention in sperm underlies rules of great generality and has a biological function. Comparing two mammalian systems we found that nucleosomes remain mainly in centromeres and in non-coding intergenic and intron regions. In contrast, coding DNA, promoter regions and transcription start and end sites, especially in homeobox genes and in the majority of genes with relevance for organ development and morphogenesis, were nucleosome-free. 146 bp mono-nucleosomal DNA was isolated and purified from sperm samples of two fertile bulls (bos taurus), one fertile man and a pool of four fertile donors; each DNA sample was deep sequenced using Illumina GAIIx and genome-wide mapped on respective genome; nucleosome-binding sites were analyzed in a genome-wide comparative manner.

Project description:Following their production in the testis, spermatozoa enter the epididymis to gain their motility and fertilizing abilities. This post-testicular maturation coincides with sperm epigenetic profile changes that influence the progeny outcome. While recent studies underscored the dynamics of small non-coding RNAs in the maturing spermatozoa, little is known regarding sperm methylation changes and their impact at the post-fertilization level. To map out the sperm methylome dynamics, we purified spermatozoa by FACS from the testis and the different epididymal segments (i.e. caput, corpus and cauda) of CAG/su9-DsRed2; Acr3-EGFP transgenic mice. Reduced-Representation Bisulfite Sequencing (RRBS-Seq) performed on DNA from these respective sperm populations indicated that high methylation changes were observed between spermatozoa from the caput vs. testis with 5546 entries meeting our threshold values (q value < 0.01, methylation difference above 25 %). Most of these changes were transitory during epididymal sperm maturation according to the low number of entries identified between spermatozoa from cauda vs. testis.

Project description:The objective of the present study is to investigate if females have the ability to recognise X or Y chromosome bearing spermatozoa and present a different response to different spermatozoa. In order to investigate if females have the ability to recognise X or Y chromosome bearing spermatozoa and present a different response to different spermatozoa. Samples of semen were sorted for chromosomal sex by M-oM-,M-^Bow cytometric-sorting. Using laparoscopic insemination one oviduct was inseminated with X spermatozoa and the contralateral oviduct was inseminated with Y spermatozoa.This allowed us to obtain samples from the oviduct in contact with X sperm and Y sperm in the the contrlateral oviduct from the same sow. Oviduct pig samples were collected for RNA isolation and hybridization on Affymetrix microarrays. Four biological replicates were performed (n= 4 sows) and a total of 8 arrays were used for microarrays study (4 arrays for oviduct samples in contact with X spermatozoa and 4 arrays for oviduct samples in contact with Y spermatozoa.

Project description:In the present study, we evaluated the alteration of protein profile of spermatozoa during the different stages of cryopreservation i.e., freshly ejaculated sperm, equilibrated sperm, and cryopreserved sperm in crossbred bulls (Bos taurus * Bos indicus). It was found that the equilibration step of cryopreservation itself caused major changes in the proteome of spermatozoa. Therefore, cryopreservation protocols should be tailored in such a way that it minimize sperm proteome alterations.

Project description:In this study, we used the iTRAQ proteomics approach to perform proteomic profiling of spermatozoa associated with sperm quality in sturgeons. The results showed 291 and 359 differentially expressed proteins in A. baerii and A. schrenckii, respectively, of which 72 were common to both species and all were upregulated in high quality compared with low quality samples. The differentially expressed proteins were mainly categorized into the generation of precursor metabolites and energy and oxidation, and they were localized to the mitochondria. Three distinguishing pathways, Arginine and proline metabolism, Pyruvate metabolism and the Citrate cycle (TCA cycle) were found to play an important role in energy metabolism, and some substrates could be used in the sperm medium for storage and cryopreservation.

Project description:G-protein coupled receptors (GPCRs) belong to the seven transmembrane receptors superfamily that in response to external stimuli, transduce signals via G proteins to initiate different intracellular signaling pathways which culminate in specific cellular responses. Although in somatic cells the GPCR participate in a broad variety of physiological processes, the expression of diverse GPCRs at the plasma membrane of human spermatozoa suggests their involvement in the regulation of sperm fertility. Mature spermatozoa could present unique features in their molecular mechanisms downstream GPCRs due to the fact that they possess sperm-specific proteins and are transcriptionally and translationally silent. In order to decipher the signaling pathways engaged by this receptor superfamily in human spermatozoa, we selected the kappa-opioid receptor (KOR) as a study model and applied, for first time, phosphoproteomic approach based on TMT labeling and LC-MS/MS analyses combined with functional studies using a specific agonist of KOR, U50488H.

Project description:The objective of the study was to identify the fertility-associated metabolites in bovine spermatozoa using liquid chromatography-mass spectrometry (LC-MS). Six Holstein Friesian crossbred bulls (three high-fertile and three low-fertile bulls) were the experimental animals. Sperm proteins were isolated and protein-normalized samples were processed for metabolite extraction and subjected to LC-MS/MS analysis. Mass spectrometry data were processed using iMETQ software and metabolites were identified using Human Metabolome DataBase while, Metaboanalyst 4.0 tool was used for statistical and pathway analysis. A total of 3,704 metabolites belonging to various chemical classes were identified in bull spermatozoa. After sorting out exogenous metabolites, 56 metabolites were observed common to both the groups while 44 and 35 metabolites were found unique to high- and low-fertile spermatozoa, respectively. Among the common metabolites, concentrations of 19 metabolites were higher in high-fertile compared to low-fertile spermatozoa (fold change > 1.00). Spermatozoa metabolites with variable importance in projections score of more than 1.5 included hypotaurine, d-cysteine, selenocystine. In addition, metabolites such as spermine and l-cysteine were identified exclusively in high-fertile spermatozoa. Collectively, the present study established the metabolic profile of bovine spermatozoa and identified the metabolomic differences between spermatozoa from high- and low-fertile bulls. Among the sperm metabolites, hypotaurine, selenocysteine, l-malic acid, d-cysteine, and chondroitin 4-sulfate hold the potential to be recognized as fertility-associated metabolites.
