We investigated the gene expression modulation in human hepatoma cell line HepG2 after exposure to 2 M-BM-5m and 10 M-BM-5m cadmium using Agilent Whole human genome expression microarray. HepG2 cells were treated for 24 hours with 2 M-NM-<M and 10 M-NM-<M Cd and normal medium as control. Three independent replicates were used for the each type of stimuli.

Project description:The trace element manganese is essential for normal development for all the organisms. Overexposure of manganese may leads to multiple neuronal disorders such as Parkinson, manganism. To explore the molecular mechanism of manganese induced neurotoxicity, gene expression profiling was performed on human neuroblastoma SHSY-5Y cells. Cells were exposed to sub-lethal concentration of manganese (100 Î¼M) for 24 hrs. Fibroblast secretome proteomic analysis

Project description:LV fibroblasts were isolated from control (day 0), and infarcted regions at day 1, day 3, and day 7 after myocardial infarction (MI). Cells were isolated in DMEM in 10% fetal bovine serum and were seeded into 6-well plates and were allowed to grow to 80% confluence in DMEM supplemented with 10% fetal bovine serum (FBS). Cells were serum starved (DMEM + 0.1% FBS) for 18 h and then media was changed and secretome was collected after 24 h. The sample sizes are n=3 biological replicates for each time point.

Global transcriptome changes in HepG2 cells exposed to copper

Project description:Our hypothesis is that copper modulates the activity of multiple intracellular signal transduction pathways to affect transcription. We have previously shown that copper activates transcription through both metal- and oxidative stress-responsive signal transduction pathways. Since the global molecular mechanisms underlying copper toxicity have not been well elucidated in humans, we have profiled transcriptome changes in HepG2 cells exposed to 100, 200, 400 and 600 uM copper for 4, 8, 12 and 24 hours using a human oligonucleotide microarray. Differentially expressed genes were identified, and integrated into biological and functional pathways through Gene Ontology analysis. Global gene expression profile was overlaid onto biomolecular interaction networks and signal transduction cascades using pathway mapping and interactome identification. Project description:This SuperSeries is composed of the following subset Series: GSE9930: Wild type 3 hour exposure to zymolyase GSE9931: hog1 mutant 3 hour exposure to zymolyase GSE9932: slt2 mutant 3 hour exposure to zymolyase GSE9933: rlm1 mutant 3 hour exposure to zymolyase GSE9934: msn2/4 mutant 3 hour exposure to zymolyase S. cerevisiae wild type (BY4741) and mutants were grown on YEPD. For Zymolyase experiments, yeast cells were grown overnight at 24 Â°C to an optical density 0.8 - 1 (A600). The culture was refreshed to 0.2 O.D and grown at 24 Â°C for 2h 30min. Next, culture was divided into two parts. One continues growing under same conditions (non-treated culture) while the other was supplemented with 5units/ml Zymolyase 100T. Cells were collected at 3 hours of growth, frozen at -80 Â°C and processed for RNA extraction. The total RNA from one culture was analyzed, and, in addition, for this sample two different hybridizations were performed, including fluorochrome swapping in order to minimize transcriptional changes due to technical variability. This therefore corresponded to two DNA microarrays analyzed.
