The expression of circular RNAs in tumor tissues from three non-cirrhotic hepatocellular carcinoma patients and three cirrhotic hepatocellular carcinoma patients

Project description:Six frozen hepatocellular carcinoma (HCC) samples (three without cirrhotic and three with cirrhotic background) were selected to analyze the circRNA expression profile through circRNA-seq. A total of 20334 circRNAs were identified in these tumor tissues, which were widely distributed in all chromosomes. Differential analysis revealed that 393 were significantly dysregulated in non-cirrhotic HCC when compared with cirrhotic HCC tissues (log2(fold change) > 1 and p < 0.05). Our study undoubtedly provide a new insight in investigating the initiation of HCC.

Trichloroethylene-induced miRNA expression changes in B6C3F1 mouse liver

Project description:To know if Trichloroethylene (TCE) can induce miRNA expression changes in mouse liver, we use microarray to screen for differentially expressed miRNAs in the liver of mice exposed to TCE at a dose of 1000 mg/kg b.w. for 5days . In our results, nine miRNAs were selected by volcano plot filtering (fold change ≥ 2 and P-value ≤ 0.05), out of which eight were up-regulated and one was down-regulated. The differential expression of miR-182-5p, miR-34a-5p and miR-451a were validated by qPCR.

The miRNA expression patterns in WSN-infected A549 cells at various time points post-infection.

Project description:To identify miRNAs that are affected by IAV infection, we adopted a systematic approach, and performed a miRNA microarray analysis using a human alveolar adenocarcinoma cell line, A549, which was infected with a common laboratory strain of influenza A/WSN/33(H1N1) virus (WSN). We compared miRNA expression profiles in A549 cells infected with WSN for 2, 6, and 10 hours at a multiplicity of infection (MOI) of 2. Among 955 miRNAs on the microarray, 209 miRNAs were detected in all three infected A549 samples, but when compared with analysis results from mock-infected A549 cells, most expression changes were less than 1.5-fold , which was consistent with a previous report (Buggele et al., 2013). Therefore, we established 1.5-fold expression change as a threshold to select for miRNAs potentially affected by WSN infection. Further screening revealed 116 miRNAs that underwent 1.5-fold or greater expression changes at any timepoint following WSN infection.

Project description:We report that ~300 mature miRNAs were detected in these cells. 73 miRNAs were overexpressed (fold change > 2; adjusted P-value < 0.05) and 14 were underexpressed (fold change > 2; adjusted P-value < 0.05) in K/VP.5 compared to K562 cells.

Relationships Between Self-Rated Health at Three Time Points: Past, Present, Future.

Project description:Background: Multiple studies have shown that people who have experienced a serious health problem such as an injury tend to overrate the quality of health they had before that event. The main objective of this study was to test whether the phenomenon of respondents overrating their past health can also be observed in people from the general population. A second aim was to test whether habitual optimism is indeed focused on events in the future. Method: A representatively selected community sample from Leipzig, Germany (n = 2282, age range: 40-75 years) was examined. Respondents were asked to assess their current health, their past health (5 years before), and their expected future health (in 5 years) on a 0-100 scale. In addition, the study participants completed several questionnaires on specific aspects of physical and mental health. Results: Respondents of all age groups assessed their health as having been better in the past than it was at present. Moreover, they also assessed their earlier state of health more positively than people 5 years younger did their current state. Habitual optimism was associated with respondents having more positive expectations of how healthy they will be in 5 years time (r = 0.37), but the correlation with their assessments of their current health was nearly as high (r = 0.36). Conclusion: Highly positive scores of retrospectively assessed health among people who have experienced a health problem cannot totally be accounted for by a response to that health problem.

RNA sequencing of LINC02015 knockdown in human aortic smooth muscle cells

Project description:This study was designed to explore the role of LINC02015 in regulating biological behaviors of vascular smooth muscle cells (VSMCs) and investigate the interacting targets of LINC02015 with high-throughput techniques and bioinformatics analysis. Total RNA was extracted from cells using Trizol Reagent (Invitrogen, Carlsbad, USA). RNA concentration was measured by NanoDrop ND-1000. RNA samples were reversely transcribed to cDNA library using the SuperScript III Reverse Transcriptase kit (Invitrogen). The gene expression profile was acquired by RNA sequencing. The differentially expressed RNAs were statistically significant with a change of >2.0 or <2.0-fold with P<0.05.
