Project description:To understand alterations in gene expression upon plakophilin3 (PKP3) loss, we generated two FBM derived plakophilin3 knockdown clones. One of these clones named shpkp3-2 was used for the experiment. The gene expression profile of shpkp3-2 was compared with the vector control named vec. RNA used for this experiment was obtained from two biological replicates of vec (vec1 and vec2) and shpkp3-2 ( shpkp3-2-1 and shpkp3-2-2 ). RNA from the clones were obtained at separate time points in 2 groups. The first group was vec1 and shpkp3-2-1 and the second group was of clones vec2 and shpkp3-2-2 respectively.

Identification of genes differentially expressed upon knockdown of CRISP-3 in human prostate cancer cells

Project description:Human prostate cancer cell line LNCaP was stably transfected with a construct expressing shRNA against CRISP-3 gene. Knockdown of CRISP-3 was confirmed at RNA and protein level in the selected stable clones at three different passages. In order to identify the possible signalling pathways regulated by CRISP-3, one of the knockdown clones was subjected to microarray analysis. Global gene expression profile of the CRISP-3 knockdown clone was compared to that of the emprty vector clone. It was observed that Kallikrein 3 was significantly downregulated upon CRISP-3 knockdown whereas Annexin A1 and Vimentin were one of the highly upregulated genes.

The study assayed the expression of 2 pairs of stably transfected colorectal cancer cell lines: The CDX1 nonexpressing CRC cell line HCT116 was stably transfected with either CDX1 cDNA in the pRC/CMV expression vector (HCT116-CDX1) or with vector control (HCT116-Vec). The CDX1-expressing CRC cell line LS174T was similarly transfected with either a pSilencer vector containing a short sequence of CDX1 siRNA (LS174T-siRNA) , or a pSilencer vector containing a scrambled siRNA sequence as a control (LS174T-Vec).

Project description:We have transfected a new tumor suppresor gene named ARLTS1 in A549 lung cancer cell line that shows a very low expression level in comparison with normal lung. This line was transfected using the pMV-7 vector containing the full length ARLTS1 coding sequence (transfected cell lines denoted FL1 to FL4), the C-terminus deleted cDNA (transfectants denoted Stop1 to Stop3) or the control (empty) vector. The expression profile was compared between the FL and Stop clones, using the wild-type as reference.
