Project description:Regulation of gene expression programs is crucial for the survival of microbial pathogens in host environments and for their ability to cause disease. Here we investigated the epigenetic regulator RSC (Remodels the Structure of Chromatin) in the most prevalent human fungal pathogen Candida albicans. We addressed the global functions of RSC in C. albicans by analyzing the changes in the proteome of cells lacking the catalytic subunit Sth1 using DIA-MS (Data Independent Acquisition-Mass Spectrometry). For this purpose, we generated a conditional mutant harboring a doxycycline (dox)-repressible allele of STH1 (sth1∆/pTetO-STH1-MYC) and treated this mutant with dox for Sth1 depletion. To assess the effect of dox alone on gene expression, a near-isogenic sth1∆/STH1-MYC strain was also analysed.

Project description:This study analyses the effects of truncation of the gene encoding Tor1 kinase in Candida albicans. The study involves a mutant tetO-TOR1-Del381 which has the N terminal regulator HEAT repeats deleted compared to a full length tetO-TOR1 strain and a heterozygous TOR1/tor1 control. Cell were grown in vitro in YEPD broth at 30˚C. In shut off experiments, doxycycline (30 µg.ml) was added to cultures to shut off the tetO promoter and inhibit Tor1 expression.

Project description:Candida albicans is an opportunistic yeast pathogen that causes a wide range of infections especially amongst immunocompromised patients. Aureobasidin A (AbA) has been shown to inhibit inositolphosphoryl ceramide synthase (IPCS), a key enzyme responsible for sphingolipid biosynthesis. There are limited studies exploring IPCS as a target molecule for antifungal treatment. It is hypothesized that the mechanism of AbA inhibition involves alteration of C. albicans phospholipid and sphingolipid profiles. The profiling of C. albicans phospholipid and sphingolipid upon exposure to 0.5-4 µg/ml of AbA were determined using Liquid chromatography-mass spectrometry (LC-MS).

Project description:Differential expression between monosoic derivative and parental strain of Candida albicans. The important human pathogen Candida albicans possesses an unusual form of gene regulation, in which the copy number of an entire specific chromosome or a large portion of a specific chromosome changes in response to a specific adverse environment, thus, assuring survival. In the absence of the adverse environment, the altered portion of the genome can be restored to its normal condition. One major question is how C. albicans copes with gene imbalance arising by transitory aneuploid states. Here, we compared transcriptomes from two copies of chromosome 5 (Ch5) in a normal diploid strain 3153A and from a single copy of Ch5 in representative derivative Sor55. Statistical analysis revealed that at least 40% of transcripts from the monosomic Ch5 are fully compensated to a disomic level, thus, indicating the existence of a genome-wide mechanism maintaining cell homeostasis. However, a minor portion of transcripts diminished twofold in accordance with what would be expected for Ch5 monosomy. Another minor portion of transcripts, unexpectedly, increased up to twofold and higher then the disomic level, demonstrating indirect control by monosomy. We suggest that C. albicans unusual regulation of gene expression by the loss and gain of entire chromosomes is coupled with widespread compensation of gene dosage at the transcriptional level.

Project description:Transcriptional profiling of CDC53 down-regulated Candida albicans cells compared to control cells Keywords: comparative genomic hybridization, genetic modification Candida albicans is an important opportunistic human fungal pathogen, which can cause mucosal as well as systemic infections in immunocompromised patients. Critical for the virulence of C. albicans is its ability to undergo a morphological transition from yeast to hyphal growth mode. Proper induction of filamentation is dependent on the ubiquitination pathway, which targets proteins for proteasome-mediated protein degradation or activates them for signaling events. In the present study, we evaluated the role of ubiquitination in C. albicans by impairing the function of the major ubiquitin-ligase complex SCF. This was done by depleting its backbone, the cullin Cdc53p (orf19.1674), using a tetracycline down-regulatable promoter system. Cdc53p-depleted cells displayed an invasive phenotype and constitutive filamentation under conditions favouring yeast growth mode, both on solid and in liquid media. In addition, these cells exhibited an early onset of cell death, as judged from propidium iodide staining, suggesting that CDC53 is an essential gene in C. albicans. To identify Cdc53p-dependent pathways in C. albicans, a genome-wide expression analysis was carried out that revealed a total of 425 differentially expressed genes (fold change ⥠2, p-value ⤠0.05) with 192 up- and 233 down-regulated genes in the CDC53-repressed mutant as compared to the control strain. GO term analysis identified biological processes significantly affected by Cdc53p depletion, including amino acid starvation response, with 14 genes being targets of the transcriptional regulator Gcn4p, and reductive iron transport. These results indicate that Cdc53p enables C. albicans to adequately respond to environmental signals.
