ABSTRACT: Susceptible (DBA/2J, 129/SvImJ, A/J) and Resistant (SM/J, C57BL/6J, Balb/cJ) mouse strain were inoculated with a highly pathogenic H5N1 influenza A virus (A/Hong Kong/213/2003) for 72 hours or not infected (control animals). Differences in expression were analyzed and used to identify candidate genes and pathways that contributed to the difference in H5N1 pathogenesis in these two groups of mice.

Project description:Susceptible and Resistant mouse strain, e.g. DBA/2J and C57BL/6J respectively, were inoculated with a highly pathogenic H5N1 influenza A virus (A/Hong Kong/213/2003) for 72 hours. Differences in expression were analyzed and use to identify candidate genes and pathways that contributed to the difference in H5N1 pathogenesis in these two strains. Recombinant inbred BXD strains are derived from the DBA/2J and C57BL/6 parent and were used to identify genetic loci associated with resistant to H5N1 infection. Female 6-8 weeks old animals were inoculated with H5N1 virus or not and 72 hours later the lungs were obtained and immediately homogenized in Trizol. The extracted RNA was submitted for Affymetrix Gene expression arrays. Differences in gene-expression were determined and used to identify candidate genes and pathways that are important for disease severity and susceptiblity to H5N1 virus.

Transcription profiling of effect of genetic background on the immune response to streptococcal infection, eight genetically diverse strains of mice (129/SvImJ, A/J, Balb/cJ, C3H/HeJ, C57BL/6J, Cast/EI, DBA/2J, and FVB/NJ)

Project description:To evaluate the effect of genetic background on the immune response to streptococcal infection, eight genetically diverse strains of mice (129/SvImJ, A/J, Balb/cJ, C3H/HeJ, C57BL/6J, Cast/EI, DBA/2J, and FVB/NJ) were infected with three doses of Streptococcus zooepidemicus (500, 5,000, or 50,000 colony forming units - CFUs) by orapharyngeal aspiration. Bacterial clearance was determined by plating out dilutions of homogenized lung tissue and counting the colonies formed after 24 hours of incubation. The mean log10 CFU per ml was calculated for each strain at all timepoints. There was a range of susceptibility between the nine strains at most doses and timepoints (6, 24, and 96 hours). At the lowest dose, the 129/SvImJ and C3H/HeJ strains had significantly higher bacterial counts than five of the other strains at both 24 and 96 hours post infection. In contrast, A/J mice cleared all of the low dose inoculum from their lungs at 96 hours. At the medium dose (5,000 CFUs), the 129/Svlm and the C3H/HeJ continue to be the most susceptible strains. The DBA/2J and the C57BL/6J were the only two strains to show an overall decrease in streptococcal growth from 6 to 96 hours post infection. Following inoculation with the highest dose of Streptococcus, the Balb/cJ strain was significantly more resistant than most other strains at the 6 and 24 hour time points. However, there were no significant differences between strains at the 96 hour time point. The results thus far demonstrate significant host differences in lung response to S. zooepidemicus that strongly suggest a genetic basis for the lung response to bacterial infections.

Transcriptomic Analysis of the Hippocampus from Six Inbred Strains of Mice Suggests Basis for Sex-Specific Susceptibility and Severity of Neurological Disorders

Project description:Identifying sex differences in gene expression within the brain is critical for determining why multiple neurological and behavioral disorders differentially affect males and females. Several are more common or severe in males (e.g., autism and schizophrenia) or females (e.g., Alzheimer’s disease and depression). We analyzed transcriptomic data from the mouse hippocampus of six inbred strains (129S1/SvImJ, A/J, C57BL/6J, DBA/1J, DBA/2J and PWD/Ph), to provide a perspective on differences between male and female gene expression. Our data show that: 1) significant gene expression differences in males versus females varies substantially across the strains, 2) 12 genes exist that are differentially expressed across the inbred strains (termed core genes), and 3) there are >2,600 significantly differentially expressed genes (DEGs) among the strains (termed non-core genes). We found that DBA/2J uniquely has a substantial majority (89%) of DEGs that are more highly expressed in females than males; 129/SvImJ is the most strongly male-biased with a majority (69%) of DEGs that are more highly expressed in males. To gain insight into the sex-biased DEGs, we examined gene ontology, pathway and phenotype enrichment and found significant enrichment in phenotypes related to abnormal nervous system morphology and physiology, among others. In addition, several pathways are enriched significantly, including Alzheimer’s disease (AD), with 32 genes implicated in AD, 8 of which are male-biased. Three of the male-biased genes have been implicated in a neuroprotective role in AD. Our transcriptomic data provide new insight into understanding the possible genetic bases for sex-specific susceptibility and severity of brain disorders.
