We compared levels of gene expression in 6-month old mice to begin to identify mechanisms of oxidative-stress induced neurodegeneration and to correlate the cellular changes that we observed in the olfactory epithelium by using histology and immunohistochemistry with gene expression changes.

Project description:Expression profiling of mRNA abundance in the adult mouse olfactory epithelium during replacement of OSNs forced by the bilateral ablation of the olfactory bulbs. The experiment was done on 6 week old male C57Bl/6 mice. Olfactory epithelium tissue samples were collected on days 1, 5, and 7 after bulbectomy. The cellular processes activated by bulbectomy include apoptosis of mature olfactory sensory neurons, infiltration of macrophages and dendritic cells, stimulation of proliferation of basal cell progenitors, and differentation of new sensory neurons.

Expression data from Emx2 wildtype and knockout olfactory epithelium

Project description:Emx2 is a homeobox transcription factor that plays a critical role in development. Olfactory sensory neuron axons from Emx2 knockout mice fail to innervate their target tissue, the olfactory bulb. Homeobox transcription factors may also play an important role in olfactory receptor expression. We used microarrays to analyze differences in mRNA abundance in the olfactory epithelium of Emx2 wildtype and knockout embryonic day 18.5 mice. Keywords: Knockout-Wildtype comparision Total RNA from 9 Emx2 wildtype and 9 Emx2 knockout mice was analyzed. Equal amounts of RNA from 3 animals was pooled to create one biological sample, 3 pools per genotype, 6 chips total. RNA was hybridized to the Affymetrix Mouse Exon 1.0 ST array.

Inflammation and degeneration of the olfactory epithelium in an immuno-deficient zebrafish

Project description:Vertebrates are protected from pathogens by an adaptive and an innate immune system. In mammals, loss of the adaptive immune system severely compromises viability. Zebrafish, in contrast, can survive for relatively long without adaptive immunity, as demonstrated by the immuno-deficient Rag1 mutant. Here, we examine whether the innate immune system is activated in the mutant, and ask whether this has any effect on the nervous system, given the finding that inflammation is linked to neurodegeneration in higher vertebrates. Using microarray analysis of the olfactory epithelium, we show that innate immune responses are upregulated. Neuronal genes are down-regulated, and immunofluorescence for the neural marker HuC shows a progressive loss of olfactory sensory neurons in mutants. Propodium iodide uptake indicates a corresponding increase in cell death. Rag1 mutant fish progressively lose their sensitivity to an olfactory cue, the alarm pheromone Schreckstoff, indicating that neuro-degeneration has a behavioral significance. Taken together, these data establish that immuno-deficiency in the zebrafish is accompanied by neuro-inflammation and loss of neurons. The microarray data document transcriptional changes associated with activation of innate immunity in the context of immunodeficiency and also identify novel genes that are potentially related to function of the olfactory system. Keywords: analysis of mutant A total of 6 RNA samples were used for hybridization. 3 were from the olfactory rosettes of wild type adult zebrafish and 3 were from Rag1 mutants.
