Project description:Analysis of gene expression profile in peritoneal macrophage extracted from LPS or PBS challenged DUSP3-/- and WT mice. DUSP3 deletion protects mice from sepsis and endotoxemia. We performed a microarray analysis to get insights into the differentially regulated pathways between WT and KO under inflammatory conditions. Total RNA obtained from isolated peritoneal macrophages isolated from DUSP3-/- and WT mice 36 hours after challenging them in vivo with PBS or with 6mg/kg of LPS.

Project description:Severe sepsis and septic shock, which occur with high incidence and are prevalent in the emergency department setting, are among the most common causes of death in hospitalized patients worldwide, and there has been a continuous increase in the incidence of sepsis of approximately 5–10% per year. Project description:Primary peritoneal macrophages were obtained from the peritoneal exudates of mice that were pretreated with 3% thioglycolate (LA4590, Solarbio) i.p. for 5 days. The peritoneal exudate cells were collected by cold PBS and adjusted to 1 × 10^6 cells/ml in DMEM cultured at 37C and 5% CO2 in a cell incubator. Casp8- and Casp11-dependent gene expression in ileal mucosa of LPS-challenged male mice

Project description:The execution of shock following high dose E. coli lipopolysaccharide (LPS) or bacterial sepsis in mice required pro-apoptotic caspase-8 in addition to pro-pyroptotic caspase-11 and gasdermin d. Hematopoietic cells produced MyD88- and TRIF-dependent inflammatory cytokines sufficient to initiate LPS shock independent of caspase-8 or caspase-11. Both proteases had to be present to support tissue injury, dependent upon TNF and interferon , first observed in the small intestine and later in spleen and thymus. Caspase-11 enhanced the activation of caspase-8 and extrinsic cell death machinery within the lower small intestine. Neither caspase-8 nor caspase-11 was individually sufficient for shock, but collaborated to amplify the inflammatory signals associated with tissue injury. Therefore, combined pyroptotic and apoptotic signaling mediated endotoxemia independent of RIPK1 kinase activity and RIPK3 function. These observations bring to light the relevance of tissue compartmentalization during inflammatory signaling in vivo where cytokines execute diverse cell death pathways and tissue damage.

High-throughput RNA sequencing analysis of mouse peritoneal cells from wild type and Lactoferrin knockout mouse during Lipopolysaccharide(LPS) stimulation

Project description:Purpose: The goal of this study is using RNA sequencing analysis to evaluate the role of lactoferrin in the LPS-induced acute inflammation in mouse Methods: 4 hs after intraperitoneal injection with LPS(4mg/kg) or equal volume PBS , the peritoneal cells were isolated from the peritoneal lavage fluid from wild type and lactoferrin knockout mice (n=3 for each group) . Then the cell RNA was sequenced by the HiSeq 2500 high-throughput sequencing service (Oebiotech, Shanghai, China). Data were extracted and normalized according to the manufacturer’s standard protocol. Results: Log-fold changes of up- or down-regulated mRNAs between the control and experiment group were selected with a significance threshold of p<0.05. Conclusions: Our study describes the mRNA expression profile changes of peritoneal cells from wild type and lactoferrin knockout mice during LPS-induced acute inflammation
