Lysolecithin as feed additive enhances collagen expression and villus length in the jejunum of broiler chickens

Project description:Adding lysolecithin to feed has reportedly improved the performance of broiler chickens. Lysolecithin is generated by phospholipase catalyzed hydrolysis of lecithin. The enzymatic reaction converts phospholipids into lysophospholipids, with lysophosphatidylcholine (LPC) the primary product. Here we compared supplementation with a commercial lysolecithin (Lysoforte(R) Kemin Industries, Inc., Des Moines, IA) with comparable levels of purified LPC for effects on broilers. Despite no differences in weight gain during the starter period, we discovered a significant increase in average villus length in the jejunum with lysolecithin, but not with LPC. High-throughput gene expression microarray analyses revealed many more genes were regulated in the epithelium of jejunum by lysolecithin compared to LPC. The most upregulated genes and pathways were for collagen, extracellular matrix and integrins. Staining sections of jejunum with Sirius Red confirmed the increased deposition of collagen fibrils in villi of broilers fed lysolecithin but not LPC. Thus, lysolecithin elicits gene expression in the intestinal epithelium leading to enhanced collagen deposition and villus length. LPC alone as a supplement does not mimic these responses. Feed supplementation with lysolecithin triggers changes in the intestinal epithelium with the potential to improve overall gut health and performance.

Skin transcriptome profiles associated with skin color in chickens

Project description:This work was to study the transcriptome profiles in the skin of chickens with black versus white skin using high-throughput RNA deep-sequencing technology, to investigate the different expression profiles of the genes involved in skin pigmentation, then look for the main differences between black and white skin colors in Lueyang chickens. 16-week-old white and black female Lueyang chickens (5 birds per color) were selected for the sample collection. A piece of skin (8 mm in diameter) from the left back was collected . Total RNA was extracted from the sample using Trizol reagent . Three RNA samples from either the black or white skin samples were pooled following mRNA isolation. The sequencing of the library was performed using an Illumina HiSeq 2000 (LianChuan Sciences, Hangzhou, China). According the result of sequencing, some colored gene expressions were validated using Real time quantitative polymerase chain reaction (qPCR).

Project description:Meat type chickens have limited capacities to cope with high environmental temperatures, sometimes leading to important mortality in farms and subsequent economic loss. One of the strategies to increase the sustainability of broiler chicken production is to enhance their adaptive capacities to face heat exposure using heat conditioning (C) during embryogenesis. This strategy was shown to improve thermotolerance in later life. The aim of this study was to determine the effect of C (39.5°C, 12h/24 vs 37.8°C from d7 to d16 of embryogenesis) and of a subsequent heat challenge (32°C for 5h, CH) applied on d34, on global gene expression in the Pectoralis major muscle (PM). Chicken Genome Array (60 K) was used to compare muscle gene expression profiles of Control (T) and heat-conditioned C chickens (characterized by low body temperature) reared at 21°C, and reared at 32°C (TCH and CCH respectively) in a dye-swap design with four comparisons and 8 broilers per condition. Real-time RT-PCR was subsequently performed to validate differential expressions in each comparison. Gene ontology, clustering and network building strategies were used.
