Comparison of transcriptomes in class-B GATA mutants with cytokinin treatments

ABSTRACT: We have used Agilent whole genome arrays (v4) to compare the gene expression changes between 14 days-old Arabidopsis seedlings with or without cytokinin treatments in wild type as well as the following mutant genotypes: gnc gnl (AT5G56860, AT4G26150), gata16 gata17 gata17l (AT5G49300, AT3G16870, AT4G16141), gnc gnl gata16 gata17 gata17l (AT5G56860, AT4G26150, AT5G49300, AT3G16870, AT4G16141) Three independent biological samples were prepared for each genotype. Experiment was performed in two rounds: Col-0_1, gnc gnl, quintuple and Col-0_2, triple.

Project description:GATA transcription factors are involved in multiple processes in plant growth and development. Two GATA factors, nitrate-inducible, carbon metabolism-involved (GNC) and CYTOKININ-RESPONSIVE GATA FACTOR 1 (CGA1, also named GNL) are important regulators in greening, flowering, senescence and hormone signaling. However, their direct target genes in regulating these biological processes are poorly characterized. Here, we report a genome-wide identification of the target genes of Arabidopsis GNC and CGA1.

Project description:To investigate the role of LLM-domain B-GATAs in starch metabolism we performed gene expression profiling analysis of RNA-seq data, obtained with Illumina technologies. B-GATA double mutant gnc gnl and the transgenic line overexpressing GNL were grown in dark conditions to avoid interference from photosynthesis or other light-dependent processes affecting sugar and starch biosynthesis.

Project description:To investigate the role of AGO10 in the buffering of cytokinin responses we grew the ago10-1 mutant and the Col-0 wildtype on plates for six daysbefore treting with mock solution or cytokinin for 2 hours. Then root tips excised and RNA was extracted using a commercial kit.
