Project description:We carried out the analysis using human skin fibrobllast HCA2 (MJ90) cells. Cells were indcued to be senescent cells by treatment with RO3306 and Nutlin3a. The transcriptome analysis between young cells (uniduced cells) nd senescent cells revealed lonrf2, a ubiqutin E3 ligase, is one of the genes highly expressed in senescnet cells.

Single cell RNA-seq of Nutlin3a-Induced Senescent mouse embryonic fibroblasts with or without expressing OSKM

Project description:Purpose: Use scRNA-seq to determine the trajectories of transcriptomes during partial reprogramming of nutlin-3a senescent MEFs. Methods: MEFs were isolated from p16-CreERT2/rtTA3-mKate2/TRE-OSKM-mCherry mice. After the senescence induction through RO3306 and nutlin-3a, treatments with 4-OHT for 9 days and doxycycline for 12 days were performed to induce OSKM expression. The doxycycline treated MEFs and untreated MEFs were collected by trypsinization and directly used for scRNA library preparation through droplet system.

Actin in young and senescent fibroblasts.

Project description:Double labelling and the isolation of peptides specific to muscle actin were used to determine the amount of actin in young and senescent fibroblasts from chicken embryo. A 20-residue peptide produced from the C-terminus of muscle actin was found in amounts that indicated 6.9% of the protein of young fibroblasts and 11.5% of the protein of senescent fibroblasts to be actin. Two other peptides of muscle actin were present in very small amounts. This indicates that the actin of both young and senescent fibroblasts is homologous to, but not identical with, muscle actin. Increased actin content of senescent fibroblasts may be related to their loss of proliferative capacity and low cell density at confluence.

Project description:Investigation of DNA methlation changing in thymidine treatment-induced MEFs. Mouse embryonic fibroblasts (MEFs) were obtained at E14.5 embryos and supplied with Dulbecco’s modified Eagle’s medium (DMEM) (Invitrogen) with 15% FBS, 100 μg/ml streptomycin and 100 U/ml penicillin. Potential hematopoietic progenitor cells (CD45+CD41+CD31+c-Kit+) were negatively selected with Biotin labeled anti-CD45, anti-CD31, anti-CD41 and anti-c-Kit antibodies by magnetic beads. Hereafter, treated MEFs are referred to as 4neg MEFs for initiation of induction. 4neg MEFs were seeded on mitomycin (MMC)-treated MEFs as feeder cells and cultured in DMEM with 15% FBS, 100 μg/ml streptomycin, 100 U/ml penicillin, 2 mM L-glutamine, 0.1 mM non-essential amino acids and 100 μM thymidine as induction medium for 2 days followed with analysis
