Gene expression signatures for Hib conjugate vaccine

Project description:To development of our gene expression, we have employed whole rhesus monkey genome microarray expression profiling as a discovery platform to identify genes with the potential to induce immune response. The peripheral blood from rhesus macaques, who were immunized in groups of three with the capsular polysaccharide (CPS antigen), carrier protein tetanus toxoid (TT) or conjugate vaccine via intramuscular injection (i.m.) in the anterolateral thigh on days 0, 30 and 60 using the formulations, were obtained on days 0, 30, 60 and 90. PBMCs were collected for microarray assays. Dynamic expression variations of eight genes (KLRC1, LGALS13, LTB4DH, NUAK1, VNN2, GALNT3, LOC710050 and LOC716305) were maintained in Hib conjugate vaccine group throughout experiment comparing with the CPS antigen group and carrier protein TT group.

Project description:This series represents HDEC cultured in 10% control sera vs. SSc sera with ACA vs. SSc sera with SCL70 Samples were analyzed at 1, 6, 12, and 24 hours The data that have been provided in the GEO Samples (GSMs) are from QuantArray. The values indicated under the VALUE header were obtained by taking the log (base 2) of the ratio of the raw data from the experimental samples to the reference RNA samples. These raw data were subsequently analyzed by Cluster and TreeView which normalizes the data across samples (see appended Table). The normalization procedure was as follows: Background corrected intensity expression levels were normalized per channel using the software program called Cluster as follows: one cycle of log transformation, five cycles of median centering of genes, and five cycles of normalizing genes to generate a log-transformed, median polished, and normal (magnitudes close to 1.0) dataset. Transcription profiling of murine macrophage-like RAW 264.7 cells treated with EdTx, protective antigen, or LPS for different length of time to examine the effect of EdTx on global transcriptional profiles.

Project description:Edema toxin (EdTx), which is a combination of edema factor and a binding moiety (protective antigen), is produced by Bacillus anthracis, the etiological agent of anthrax. EdTx is an adenylyl cyclase enzyme that converts adenosine triphosphate to adenosine-3',5'-monophosphate, resulting in interstitial edema seen in anthrax patients. We used GeneChip analysis to examine global transcriptional profiles of EdTx-treated RAW 264.7 murine macrophage-like cells at 3 and 6 hr. Experiment Overall Design: RAW 264.7 cells were treated with EdTx (2.5 µg/ml of protective antigen and 0.625 µg/ml of Edema factor), PA (2.5 µg/ml), or LPS (1 ng/mL) for 0, 3, and 6 hr. Each experiment was performed in triplicate, generating a total of 21 arrays (biological replciates).
