RNA sequencing of epidermal stem cells in Fbln7 WT vs Fbln7 KO mice

Project description:This study was performed to compare the transcriptome of epidermal stem cells in young (2 months old) and middle aged (1 year old) fibulin 7 wild type (WT) compared to fibulin 7 knockout (KO) mice. Fbln7 gene was an ECM gene significantly upregulated in aging fast-cycling stem cells and this RNA-sequencing was done to investigate the role of Fbln7 in aging epidermal stem cells. Data indicates differences in gene expressions occuring between WT and KO at 1 year old but not at 2 months old, suggesting an age dependent role of fibulin 7. At 1 year old, other matrix-related and remodelling genes were affected, together with inflammatory responses of stem cells and lineage fate abberations, as in the case of 2 years old fast-cycling epidermal stem cells.

Altered fate of tendon-derived stem cells isolated from a failed tendon-healing animal model of tendinopathy.

Project description:We hypothesized that altered fate of tendon-derived stem cells (TDSCs) might contribute to chondro-ossification and failed healing in the collagenase-induced (CI) tendon injury model. This study aimed to compare the yield, proliferative capacity, immunophenotypes, senescence, and differentiation potential of TDSCs isolated from healthy (HT) and CI tendons. TDSCs were isolated from CI and healthy Sprague-Dawley rat patellar tendons. The yield, proliferative capacity, immunophenotypes, and senescence of TDSCs (CI) and TDSCs (HT) were compared by colony-forming unit assay, BrdU assay, flow cytometry, and ?-galactosidase activity assay, respectively. Their osteogenic and chondrogenic differentiation potentials and mRNA expression of tendon-related markers were compared using standard assays. More TDSCs, which showed a lower proliferative potential and a higher cellular senescence were present in the CI patellar tendons compared to HT tendons. There was a higher alkaline phosphatase activity and mineralization in TDSCs (CI) in both basal and osteogenic media. More chondrocyte-like cells and higher proteoglycan deposition, Sox9 and collagen type II expression were observed in TDSCs (CI) pellets upon chondrogenic induction. There was a higher protein expression of Sox9, but a lower mRNA expression of Col1a1, Scx, and Tnmd in TDSCs (CI) in a basal medium. In conclusion, TDSCs (CI) showed altered fate, a higher cellular senescence, but a lower proliferative capacity compared to TDSCs (HT), which might contribute to pathological chondro-ossification and failed tendon healing in this animal model.

Transcriptome analysis of LT-HSC isolated from WT and CD169DTR/+ mice 5 days after a 2 Gy-Total Body Irradiation

Project description:A 2 Gy-total body irradiation (TBI) of WT mice induces a second wave of ROS and a second wave of apoptosis in hematopoietic stem cells starting from 6 days after irradiation. In CD169DTR/+ mice, which are depleted in CD169+ resident macrophages after a diphtheria toxin injection, these two waves do not occur and a total recovery of the LT-HSC pool is observed, contrarily to the LT-HSC pool from WT mice, which is decreased. Our goal is to understand why these secondary waves of apoptosis and ROS occur in LT-HSC from irradiated WT mice and not in LT-HSC from irradiated mice depleted in resident macrophages, allowing a recovery of the LT-HSC pool. We used microarrays to compare the global gene expression in LT-HSC isolated from WT and CD169DTR/+ mice 5 days after irradiation, i.e. just before both secondary waves of ROS and apoptosis occurred in WT mice.
