Project description:Dietary amino acids restriction extends lifespan in diverse species ranging from flies to mammals. The evolutionarily conserved serine/threonine kinase General Control Nonderepressible 2 (GCN2) is a key sensor of amino acid deficiency and has been implicated in lifespan regulation upon dietary restriction. However, the role of individual essential amino acids (EAA) in modulating organismal lifespan and the underlying molecular mechanisms through which EAA mediate these effects are only partially understood. We generated a novel Drosophila GCN2 null mutant and systematically analyzed its response to individual amino acid deficiency.

Cell-type-specific modulation of longevity by Forkhead family transcription factors in the Drosophila nervous system

Project description:We previously found that the Drosophila FoxA transcription Fork Head (FKH) is necessary for reduced insulin signalling to extend lifespan. We have now identified that increased expression of FKH in neurons alone is sufficient to extend healthy lifespan. We carried out RNA-sequencing in head tissue on controls (daGS>InRDN,GFP-RNAi without RU-486) and reduced insulin signalling with control RNAi (daGS>InRDN,GFP-RNAi with RU486) and reduced insulin signalling with FKH RNAi (daGS>InRDN,FKH-RNAi with RU486). InR-DN stock used: K1409A.

The effect of larval diet on the fecundity-longevity relationship and age-related gene expression in Drosophila melanogaster

Project description:Our two main aims were 1) to isolate age-related changes in gene expression in females of the solitary insect, Drosophila melanogaster; and 2) to determine whether experimentally changing the shape of the fecundity-longevity relationship (by changing larvae diet) caused changes in age-related gene expression in these females. To address these aims we extracted RNA from three key tissues (fat body, head and ovary) from females at two time points (10% and 60% mortality phases). Each of these females had experienced one of two treatments: medium-quality larval diet (M) treatment (resulting in a positive fecundity-longevity relationship) and high-quality larval diet (H) treatment (resulting in a negative fecundity-longevity relationship).

Chronic Circadian Misalignment Results in Reduced Longevity and Large-Scale Changes in Gene Expression in Drosophila

Project description:We investigated the consequences of chronic circadian misalignment (CCM) in the powerful model system of the fruit fly, Drosophila melanogaster. We subjected flies to daily 4-hr phase delays in the light-dark schedule and used the Drosophila Activity Monitoring (DAM) system to continuously track locomotor activity and sleep while simultaneously monitoring fly lifespan. Consistent with previous results, we find that exposing flies to CCM leads to a ~15% reduction in median lifespan in both male and female flies. Importantly, we demonstrate that the reduced longevity occurs independent of changes in overall sleep or activity. To uncover potential molecular mechanisms of CCM-induced reduction in lifespan, we conducted whole body RNA-sequencing to assess differences in gene transcription between control and misaligned flies. CCM caused progressive, large-scale changes in gene expression characterized by upregulation of genes involved in response to toxic substances, aging and oxidative stress, and downregulation of genes involved in regulation of development and differentiation, gene expression and biosynthesis. Many of these gene expression changes mimic those that occur during natural aging, consistent with the idea that CCM results in premature organismal decline, however, we found that genes involved in lipid metabolism are overrepresented among those that are differentially regulated by CCM and aging. This category of genes is also among the earliest to exhibit CCM-induced changes in expression, thus highlighting altered lipid metabolism as a potentially important mediator of the negative health consequences of CCM.

Project description:Understanding the molecular mechanisms underlying variation in lifespan is central to ensure long life and postpone aging. There is growing evidence that changes in expression of key lifespan regulators during early development are crucially important for lifespan determination.To study the early developmental impact of Lim3on Drosophila lifespan we performed a Lim3knockdown at early stages of Drosophiladevelopment. We detected the significant decrease of lifespan both in Drosophila males and females with depleted Lim3expression and revealed it to be associated with mitochondriala activity.
