A Long-Range cis-Regulatory Element for Class I Odorant Receptor Genes

ABSTRACT: Individual olfactory sensory neurons express a single odorant receptor (OR) gene from either class I genes residing in a single cluster on a single chromosome or class II genes spread over multiple clusters on multiple chromosomes. Here, we identify an enhancer element for mouse class I genes, the J element, that regulates class I gene expression of a much larger number of genes and over a much greater genomic distance than known class II gene enhancers. Our data reveal an extraordinary long-range cis-regulatory element that governs the singular class I gene expression and has been phylogenetically preserved to retain a single cluster organization of class I genes in mammals.

Effects of Bcl11b-deficiency on gene expression in the main olfactory epithelium of neonatal mice

Project description:Each olfactory sensory neuron (OSN) chooses a single odorant receptor (OR) gene from the class I or class II OR repertoire in mice. We have found that the transcription factor Bcl11b, which is coexpressed with class II OR genes both in frog and mouse, determines the class of OR to be expressed in OSNs. Both loss- and gain-of-function analyses demonstrate that Bcl11b is necessary for class II genes expression, by suppressing the class I-specific enhancer. Overall, we identified a unique transcriptional mechanism that serves as a binary switch for the OR class choice that is likely to have contributed to the acquisition of terrestrial-specific class II OR expression.

Translation Error Clusters Induced by Aminoglycoside Antibiotics

Project description:Aminoglycoside antibiotics (AGAs) target the ribosome and induce mistranslation, yet which translation errors induce bacterial cell death is unclear. The analysis of cellular proteins by quantitative mass spectrometry shows that bactericidal AGAs induce not only single translation errors, but also clusters of errors in full-length proteins in vivo with as much as four amino acid substitutions in a row. The downstream errors in a cluster are much more frequent than the first error (0.4 vs. 10-3, respectively) and independent of the intracellular AGA concentration. The prevalence, length, and composition of error clusters depends not only on the misreading propensity of a given AGA, but also on its ability to inhibit ribosome translocation along the mRNA. Error clusters constitute a new class of misreading events in vivo that may constitute the predominant source of proteotoxic stress at low AGA concentration, which is particularly important for the autocatalytic uptake of the drugs.

Biosynthesis of Isonitrile Lipopeptides by Conserved Non-ribosomal Peptide Synthetase Gene Clusters in Actinobacteria

Project description:A putative lipopeptide biosynthetic gene cluster is conserved in many species of Actinobacteria, including Mycobacterium tuberculosis and Mycobacterium marinum, but the specific function of the encoding proteins has been elusive. Using both in vivo heterologous reconstitution and in vitro biochemical analyses, we have revealed that the five encoding biosynthetic enzymes are capable of synthesizing a new family of isonitrile lipopeptides (INLPs) through a thio-template mechanism. The biosynthesis features the generation of isonitrile from a single precursor Gly promoted by a thioesterase and a non-heme iron(II)-dependent oxidase homologue, and the acylation of both amino groups of Lys by the same isonitrile acyl chain facilitated by a single condensation domain of a non-ribosomal peptide synthetase (NRPS). In addition, the deletion of INLP biosynthetic genes in M. marinum has decreased the intracellular metal concentration, suggesting the role of this biosynthetic gene cluster in metal transport.
