Transcription profiling of mouse early skin development

Project description:Mice with the spontaneous uncv mutation are hairless. We utilize the uncv null mouse to investigate the underlying mechanisms giving rise to the relevant phenotypes and reveal significant changes in certain signaling pathways by microarray. Experiment Overall Design: Dorsal skins were taken from 4-day-old uncv null mice and wild type mice (as control), and they are sisters-german. The parent is uncv heterozygosis(uncv +/-), and their children has 3 genotype: wild(uncv +/+), heterozygosis(uncv +/-) and null (uncv -/-). We can easily distinguish them by their beard length and shape or identify their genotype by PCR using specific primers. HR 02 is a wild mouse sample and uncv null mice expresses as HR 03 and HR 04. RNA isolation was performed with the QIAGEN RNeasy mini kit (QIAGEN, Valencia, CA, USA) following standard instructions.

Project description:Study to optimize our protocol for isolating RNA from skin biopsies from (hairless) SKH mice using different-diameter biopsy punches. Some mice were also treated with UVB radiation to check its effect on RNA yield. 4 mice total: 2 were irradiated with 300J/m2 UVB and 2 were non-irradiated. Post-mortem skin biopsies with 1.5mm, 2.0mm, and 2.5mm diameter punches were taken from the dorsal region.

Maternal DNA methylation regulates early trophoblast development

Project description:Critical roles for DNA methylation in embryonic development are well established, but less is known about the roles of DNA methylation during trophoblast development, the extraembryonic lineage that gives rise to the placenta. Here we dissected the role of DNA methylation in trophoblast development by performing mRNA and DNA methylation profiling of Dnmt3a/3b-null trophoblast. We find that most gene deregulation is explained by an erasure of maternal methylation in the oocyte, but partially independent of loss of imprinting of the trophoblast-essential Ascl2 gene. Unveiling complexity and multipotentiality of early heart fields

Project description:Through high-resolution single cell and genetic lineage/clonal analyses, we show an unsuspected clonal relationship between extraembryonic mesoderm and cardiac lineages. Single-cell transcriptomics and trajectory analyses uncovered two mesodermal progenitor sources contributing to left ventricle cardiomyocytes, one embryonic and the other with an extraembryonic gene expression signature. Additional lineage-tracing studies revealed that the extraembryonic-related progenitors reside at the embryonic-extraembryonic interface in gastrulating embryos, and produce distinct cell types forming the pericardium, septum transversum, epicardium, dorsolateral regions of the left ventricle and atrioventricular canal myocardium, and extraembryonic mesoderm. Clonal analyses demonstrated that these progenitors are multipotent, giving rise to not only cardiomyocytes and serosal mesothelial cell types but also, remarkably, extraembryonic mesoderm.
