Although the use of honey bees for pollination can be considered as disruption of the habitat, its effects on honey bees' physiology have never been addressed. In Japan, more than 100 thousands colonies are annually used for pollination, and intriguingly 80% of them are used in greenhouses. Recently, honey bee colonies have often collapsed when they are introduced into greenhouses. Thus, to suppress colony collapses and maintain the number of worker bees in the colonies are essential for successful long-term pollination in greenhouses and recycling honey bee colonies.

Project description:Background The number of managed honey bee colonies has considerably decreased in many developed countries in recent years and the ectoparasitic mites are considered as major threats to honey bee colonies and health. However, their general biology remains poorly understood. Results We sequenced the genome and transcriptomes of Tropilaelaps mercedesae, the prevalent ectoparasitic mite infesting honey bees in Asia. The de novo assembled genome sequence (353 Mb) represents 53% of the estimated genome size because of the compression of repetitive sequences; nevertheless, we predicted 15,190 protein-coding genes which were well supported by the mite transcriptomes and proteomic dataes. Although amino acid substitutions have been accelerated within the conserved core genes in of two mites, T. mercedesae and Metaseiulus occidentalis, T. mercedesae has undergone the least gene family expansion and contraction between the seven arthropods we tested. The number of sensory system genes has been dramatically reduced; meanwhile, T. mercedesae may have evolved a specialized cuticle and water homeostasis mechanisms, as well as epigenetic control of gene expression for ploidy compensation between males and females., and water homeostasis. T. mercedesae contains all gene sets required to detoxify xenobiotics, enabling it to be miticide resistant. T. mercedesae is closely associated with a symbiotic bacteriuma (Rickettsiella grylli-like) and DWVdeformed wing virus (DWV), the most prevalent honey bee virus. The presence of DWV in both adult male and female mites was also confirmed by the proteomic analysis. Conclusions T. mercedesae has a very specialized life history and habitat as the ectoparasitic mite strictly dependsing on the honey bee inside the a stable colony. Thus, comparison of the genome and transcriptome sequences with those of a tick and free-living mites and tick has revealed the specific features of the genome shaped by interaction with the honey bee and colony environment. T. mercedesae, as well as Varroa destructor, genome and transcriptome sequences not only provide insights into the mite biology, but may also help to develop measures to control the most serious pests of the honey bee.

We have employed Illumina/Solexa sequencing to examine the small RNA content in the bee larval food source, and show that worker jelly is enriched in miRNA complexity and abundance relative to royal jelly. The miRNA levels in worker jelly were 7-215 fold higher than in royal jelly, and both jellies showed dynamic changes in miRNA content during the 4th to 6th day of larval development. Adding specific miRNAs to royal jelly elicited significant changes in queen larval mRNA expression and in morphological characters of the emerging adult queen bee. We propose that miRNAs in the nurse bee secretions constitute an additional element in the regulatory control of caste determination in the honey bee.

Project description:Experimental infection of (2 days old) adult honey bee workers (30 bees per replicates, 3 replicates per treatments, from 3 different colonies (one colony per cage for each treatment)) with 10^9 genome equivalent of Black Queen Cell Virus (BQCV) in 10µl of sugar solution and/or 10^5 fresh Nosema ceranae spores (control bees were given a similar bee extract in PBS, without pathogen). Bees were kept in cages of 30 bees in incubator (30°C/50%RH). At day 13 p.i., bees were flash frozen, and stored at -80°C.

Project description:The timing and amplitude of reproductive effort are central life history variables for all organisms. In social insects, reproductive effort is collectively controlled at the colony level but little is known about the mechanisms that determine how much colonies invest in reproduction. As part of their female reproductive investment, honey bee colonies raise multiple new queens by feeding royal jelly to female larvae. Artificial selection for commercial royal jelly production in China has generated over the past 40 years a stock of royal jelly bees that raises an order of magnitude more queens and provisions each queen with >3x more royal jelly than unselected stock. Here we establish in a reciprocal cross-fostering experiment that this dramatic shift in social phenotype is due to changes in the nurse bees that care for the brood. We demonstrate higher electrophysiological responsiveness to brood pheromones in royal jelly bees than in unselected bees. Comparing the antennal proteome of unselected and royal jelly bees, we identify proteins involved in chemosensation and energy metabolism as candidates for the observed differences. We confirm several candidates, most prominently OBP16 and CSP4, with quantitative differences of corresponding mRNA levels and functional binding assays between the brood pheromones and the chemosensory proteins. Furthermore, we complement analyses of brood volatiles and electrophysiological recordings with behavioral attraction assays to confirm the presumed biological function of one newly discovered and two existing larval pheromones. Together, these findings help our understanding of pheromonal communication in honey bees and explain how sensory changes in nurse honey bees as alloparental caregivers have evolved in response to artificial selection, leading to a profound shift in colony-level resource allocation to sexual reproduction.

Project description:Using the honey bee (Apis mellifera) as a model, we confirmed that honey bee queens actively adjust egg size in response to the number of workers: Queens lay smaller eggs in colonies with more worker bees than when in colonies with few workers. Reduced colony size also causes decreasing ovary weight but upregulation of energy production and protein processing in the ovaries. Complementary metabolomic and proteomic analyses of the produced eggs indicate that amino acid metabolism, cell development, and cell maturation processes are elevated in bigger eggs. Our study thus provides the first mechanistic insights into the adjustment of egg size in the honey bee, a model for complex social systems that demand integration of individual and collective life-history optimization.

We collected worker and royal jelly of the Italian honeybee (ZND No.1, Apis mellifera ligustica) at 73~90 hours (4th-day larvae), 97~114 hours (5th-day larvae), and 121~138 hours (6th-day larvae) after hatching. After total RNA was extractedM-BM- and quantified , relative equal amounts of total RNAs from each of the three sampling days were pooled into respectively worker and royal jelly samples, and the fraction of small RNAs less than 30nt long was retained and sequenced on the Illumina/Solexa high-throughput platform (HTP).

Project description:This experiment examines gene expression profiles in individual honey bee brains (adult worker Apis mellifera). The purpose is to test whether behavioral phenotype can be predicted by expression profiles in individual brains in a naturalistic context (i.e., colonies in the field). The two behavioral phenotypes examined are 'nurse' and 'forager'.
