Efficient engineering of human and mouse primary cells using peptide-assisted genome editing

Project description:Simple and efficient delivery of CRISPR genome editing systems in primary cells remains a major challenge. Here, we describe an engineered Peptide-Assisted Genome Editing (PAGE) CRISPR-Cas system for rapid and robust editing of primary cells. PAGE couples a cell-penetrating Cas protein with a cell-penetrating endosomal escape peptide in a 30-minute incubation that yields up to ~98% editing efficiency in primary human and mouse T cells. PAGE provides a broadly generalizable platform for next generation genome engineering in primary cells. Comparative genomic hybridization of Schistosoma japonicum Chinese and Philippine isolates

Project description:Chinese and Philippine strains of the blood fluke Schistosoma japonicum present clear and distinctive phenotypes in areas of fecundity, pathology, drug sensitivity and immunology. Despite these differences large scale sequencing efforts have focused solely on Chinese mainland strain of the parasite. We have undertaken a comparative genomic hybridisation (CGH) approach to highlight some of the structural differences in the genome of two of the major geographical isolates of S. japonicum. We identified seven distinct regions of the S. japonicum genome that present differential CGH between Chinese and Philippine strains of the blood fluke Schistosoma japonicum, representing either deletion or duplication regions in the Philippine strain. Within these regions, genes that may be related to phenotypical differences are identified and discussed. Genomic DNA was isolated from adult (7 week post cercarial challenge) Schistosoma japonicum Chinese and Philippine isolates and separate maleand femalesamples comparatively hybridised on an Agilent customn designed oligo microarray.

Coupland et al., Cryo-EM of synaptic vesicles reveals loading-induced V-ATPase dissociation

Project description:This dataset consists of 4 raw mass spectrometry files and associated peak lists and results files, acquired on a Bruker timsTOF Pro 2 mass spectrometer operated in Data-Dependent Acquisition mode. Synaptic vesicle and cleared rat brain lysate samples were prepared and separated in SDS-PAGE gel, then processed for in-gel protease digestion by Claire Coupland. Mass spectrometric acquisition and database searches were performed by Network Biology Collaborative Centre at the Lunenfeld-Tanenbaum Research Institute. Mass spec results were analyzed by Ji-Young Youn and John Rubinstein. The files are associated with a manuscript by Coupland et al. that determines the structure of synaptic vesicle V-ATPases and their associated proteins using Cryo-EM structural analysis. Mass spectrometry analysis is used to determine the protein components in purified synaptic vesicles.
