Gene expression profiles of normal mouse mammary cell subpopulations FACS sorted based on expression of biomarkers

Project description:To delineate epithelial subpopulations in mouse mammary tissue, hematopoietic and endothelial cells were depleted from freshly isolated cell suspensions derived from mammary glands using fluorescence-activated cell sorting. The resultant Lin- population was fractionated into four distinct subpopulations using antibodies against CD29, CD24 and CD61. Based on the immunohistochemical phenotype, and in vivo and in vitro functional assays, these subpopulations were identified as fibroblast-containing stromal (CD29loCD24-), mammary stem cell (MaSC)-enriched (CD29hiCD24+CD61+), luminal progenitor (CD29loCD24+CD61+), and mature luminal (CD29loCD24+CD61-) cell subpopulations. Microarray profiling was used to derive gene expression signatures of these 4 subpopulations. The four mammary cell subpopulations were found to have distinct gene expression profiles. Four mammary cell subpopulations from 3-5 pooled mouse tissues were analysed. MS is for the MaSC-enriched cell subpopulation. LP is for the Luminal Progenitor subpopulation. ML is for the Mature Luminal subpopulation.

Hemocyanin facilitates lignocellulose digestion by wood-boring marine crustaceans.

Project description:Woody (lignocellulosic) plant biomass is an abundant renewable feedstock, rich in polysaccharides that are bound into an insoluble fiber composite with lignin. Marine crustacean woodborers of the genus Limnoria are among the few animals that can survive on a diet of this recalcitrant material without relying on gut resident microbiota. Analysis of fecal pellets revealed that Limnoria targets hexose-containing polysaccharides (mainly cellulose, and also glucomannans), corresponding with the abundance of cellulases in their digestive system, but xylans and lignin are largely unconsumed. We show that the limnoriid respiratory protein, hemocyanin, is abundant in the hindgut where wood is digested, that incubation of wood with hemocyanin markedly enhances its digestibility by cellulases, and that it modifies lignin. We propose that this activity of hemocyanins is instrumental to the ability of Limnoria to feed on wood in the absence of gut symbionts. These findings may hold potential for innovations in lignocellulose biorefining.

Small RNA content in mouse serum upon S.mansoni infection

Project description:MicroRNAs (miRNAs) are a class of short non-coding RNA that play important roles in disease processes in animals and are present in a highly stable cell-free form in body fluids. Here we examine the capacity of host and parasite miRNAs to serve as tissue or serum biomarkers of Schistosoma mansoni infection. Sequencing of small RNAs from serum confirmed the presence of miRNAs and revealed 11 parasite-derived miRNAs that were detectable by 8 weeks post S.mansoni infection.

Histone methylation profiles of prospectively isolated mammary epithelial subpopulations

Project description:In this study we mapped H3K27me3, H3K4me3 and H3K9me2 marks in three mammary epithelial subsets: MaSC/basal (MS), luminal progenitor (LP) and mature luminal (ML) in the steady-state. In addition, profiles were generated for H3K4me3 and H3K27me3 marks in the MaSC/basal and luminal populations from the glands of ovariectomized or mid-pregnant (12.5 days) mice as well as from control virgin mice. We used ChIPseq analysis to determine histone modification marks in the MS, LP and ML subsets in the steady-state. We then determined the histone modification profiles of MS and sorted luminal (Lum) cells from pregnant (12.5 dP) or ovariectomized (OVX) mice and compared these with the profiles of control virgin mice in order to study the effect of hormones on the mammary epigenomes.

Plasma Protein Corona on Polystyrene Nanoparticles Facilitates their Transfer across the Human Placenta

Project description:Nanoparticles exposed to biological fluids are rapidly surrounded by proteins. It is known that this formed protein corona influences the interplay of nanoparticles with cells or tissue barriers. In this study, we report the impact of a formed human plasma protein corona on the transfer of 80 nm polystyrene (PS) nanoparticles across the human placenta. We used the human ex-vivo placental perfusion model, as it reflects the intact and physiological tissue barrier between mother and unborn. Our results show an enhanced transfer of polystyrenes, exposed to human plasma, across the placenta compared to bovine serum albumin which served as control setting. We isolated nanoparticles before and after tissue exposure and analyzed their protein corona via shotgun proteomics and LC-MS/MS. The corona profile of particles that crossed the placenta highlighted several proteins as possible drivers for elevated tissue transfer. Subsequently two distinct proteins, human albumin and immunoglobulin G, were selected and incubated with the nanoparticles to form a sole protein corona. Strikingly, the protein corona formed by albumin induced significantly the transfer of polystyrenes across the tissue as compared to corona formed by immunoglobulins. To conclude, our study provides a comparative analysis between different formed protein coronas on nanoparticles and a corona dependent transfer behavior of polystyrenes across placental tissue. Our findings suggest that protein corona analyses of nanoparticles might help to understand better their properties at biological barriers.

Glycosidades of Pomacea canaliculata

Project description:In this study, liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) was applied to define the occurrence, diversity and origin of glycosyl-hydrolases along the digestive tract of P. canaliculata. Cellulases, hemicellulases, amylases, maltases, fucosidases and galactosidases were identified across the digestive tract. The digestive gland and the contents of the crop and style sac yield a higher diversity of glycosidase-derived peptides.
