The left ventricular phosphoproteome in mouse with cardiac-specific adenylyl cyclase type 8 overexpression

ABSTRACT: To study the molecular mechanism of adaption in the mouse with heart-specific overexpression of Adenylyl Cyclase 8 (TGAC8), we performed TMT-labeling mass spectrometry to obtain both total proteome and phosphoproteome of mouse left ventricles. Proteins were extracted from five wild-type and four TGAC8 mice and then digested with trypsin. After TMT labeling, the labeled peptides were mixed and loaded into RP-HPLC and LC-MS/MS (Thermo Orbitrap Lumos mass spectrometer). For phosphoproteome, pepetides were first enriched with TiO2 and the flow-through from TiO2 went for Fe-IMAC enrichment. The database search and statistical analysis were conducted within Proteome Discover 2.2 software.
