Project description:Phosphorus is one of the most important macronutrients that is required for plant growth and development. However, stress under low-P conditions has become a limiting factor that affects crop yields and qualities. Plants have developed strategies to cope with this, while few genes associated with low-P tolerance have been identified in soybean. We used microarrays to detail the global programme of gene expression under different phosphorus treatments of two soybean accessions CD and YH with different phosphorus efficiency. The roots and leaves of a low-P-tolerant accession and a low-P-sensitive accession were harvested after 10 days of hydroponics under different P treatments, each with three biological replicates.Then microarray chips were performed on the 24 samples. We sought to identify genes associated with low-P stress. To that end, we analyzed the differently expressed genes between different P treatments, different accessions and different tissues.

Metabolite Profiling and Classification of Highbush Blueberry Leaves under Different Shade Treatments.

Project description:Blueberry belongs to the genus Vaccinium L. in the Ericaceae and is an economically important shrub that produces small berries that are rich in nutrients. There were differences in the appearance of blueberry leaves under different shade treatments. To explore the differences in metabolites in blueberry leaves under different shading treatments, nontargeted liquid chromatography-mass spectrometry (LC-MS) metabonomic analysis was performed. Different shade intensities resulted in significant differences in the contents of metabolites. A total of 6879 known metabolites were detected, including 750 significantly differentially expressed metabolites, including mainly lipids and lipid-like molecules and phenylpropanoid and polyketide superclass members. Based on a Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the flavone and flavonol biosynthesis pathways were the most significantly enriched. The results of this study provide a reference and scientific basis for the establishment of a high-quality and high-yield shaded blueberry cultivation system.

Genome-wide identification and expression profiles of CsHIPP family genes under heavy metal stress in Camellia sinensis (L.)

Project description:HIPP (heavy metal associated isoprenylated plant protein) is a group of metal-binding metallochaperones, which plays crucial roles in metal homeostasis and detoxification. Up to now, little information of CsHIPP family genes in C. sinensis has been reported. In present study, a total of 56 CsHIPP genes from C. sinensis were identified, which were unevenly distributed on 15 chromosomes. The CsHIPP family genes were divided into five groups based on the topology of phylogenetic tree and identified AtHIPPs and OsHIPPs. Motif and gene structure analysis showed that the members in the same group showed similar conserved motifs and intron/exon structure. The expression pattern of CsHIPP family genes in different organs of C. sinensis was investigated using the previous RNA-seq data. The expression level of CsHIPP genes in C. sinensis under Cd and As treatments was also explored based on our RNA-seq analysis. The results showed that 16 CsHIPP genes displayed the same expression pattern under Cd and As treatments, which were also verified by qRT-PCR. Among them, three CsHIPP genes (CsHIPP15, CsHIPP36, and CsHIPP4) were increased at both 10 d and 15 d after Cd and As treatments, and one gene (CsHIPP40) was increased to a maximum at 10 d, and decreased at 15 d of Cd and As treatments, indicating that these four CsHIPP genes in C. sinensis might be involved in the resistance to Cd and As stresses. These results provide a basis for the functional characterization of CsHIPP genes and investigations on the molecular mechanisms of heavy metal homeostasis in C. sinensis.

Cadmium (Cd) induced expression changes in the Arabidopsis thaliana accessions Col-0 and Bur-0

Project description:Metal tolerance is often a result of metal storage or distribution. Thus, with the goal of advancing the molecular understanding of such metal homeostatic mechanisms, natural variation of metal tolerance in Arabidopsis thaliana was investigated. Substantial variation exists in tolerance of excess copper (Cu), zinc (Zn) and cadmium (Cd). Two accessions, Col-0 and Bur-0, and a recombinant inbred line (RIL) population derived from these parents were chosen for further analysis of Cd and Zn tolerance variation, which is evident at different plant ages in various experimental systems and appears to be genetically linked. Three QTLs, explaining in total nearly 50 % of the variation in Cd tolerance, were mapped. The one obvious candidate gene in the mapped intervals, HMA3, is unlikely to contribute to the variation. In order to identify additional candidate genes the Cd responses of Col-0 and Bur-0 were compared at the transcriptome level. The sustained common Cd response of the two accessions was dominated by processes implicated in plant pathogen defense. Accession-specific differences suggested a more efficient activation of acclimative responses as underlying the higher Cd tolerance of Bur-0. The second hypothesis derived from the physiological characterization of the accessions is a reduced Cd accumulation in Bur-0. The microarray analysis was used to identify candidate genes for Cd-tolerance and -accumulation differences between the accessions Bur-0 and Col-0 as well as to analyse the expressional response of A.thaliana to Cd-stress.

Project description:This project unraveled the molecular mechanism of adaptive changes of strain PAH02 in the process of absorbing Cd and degrading Phe by Tandem Mass Tags proteomic technologies.The results show that strain PAH02 significantly upregulated the levles of proteins such as MerR and aerC to ensure energy utilization and cellular integrity under the stress of Cd-Phe co-contaminant.
