Metabolomics of Mice Cohousing and Microbiota Transfer

ABSTRACT: The mice serum samples were extracted and prepared for a combined flow injection (FIA) and LC-MS/MS assay (Biocrates AbsoluteIDQ® p180 kit). The Biocrates AbsoluteIDQ® p180 kit (Biocrates Life Sciences AG, Innsbruck, Austria) can quantitatively measure ~188 biologically relevant metabolites from five analyte groups (acylcarnitines, amino acids, biogenic amines, glycerophospho- and sphingolipids, and hexoses) on a triple quadrupole mass spectrometer.

Project description:A targeted metabolomics analysis was performed on serum samples from rats immunized with a methamphetamine-like hapten covalently bound to keyhole limpet hemocyanin (KLH) monomers, and controls. The subjects were approximately 300 g adult male Sprague Dawley rats. RTI International RCMRC received pre-immune serum from RI 11-03A (n=12) which is a matched control serum that was collected before starting immunizations, and week 17 bleeds from RI 11-03A (n=12), RI 11-03B (n=8), and RI 11-03C (n=12). The A-C nomenclature denotes A) complete antigen with adjuvant, B) complete antigen without adjuvant and C) only KLH carrier protein and adjuvant without a METH-like hapten. Project description:Metabolite concentrations were obtained in prostate cancer and case control plasma samples from AA and EA individuals using the AbsoluteIDQ kit p180 (Biocrates Life Science AG, Austria) according to the manufacturer’s instructions. The analysis was performed using a QTRAP 6500 LC/MS/MS System (AB SCIEX, USA) equipped with an electrospray ionization source, Agilent G1367B autosampler and the Analyst 1.51 software (AB SCIEX, USA).

Metabolic signature of C-protein induced myositis mouse model

Project description:Objective of this study was to find Metabolic signature of C-protein induced myositis mouse model. For metabolite quantification, we used the tissue samples of C-protein induced myositis mouse model and control. Biocrates p180 kit enables to quantify 188 metabolites including amino acids, biogenic amines, acylcarnitine, glycerophospholipid and sugar. Significant metabolites were selected by t-test with false discovery rate p-value less than 0.05. In mouse tissue sample, a total of 68 metabolites significantly changed in CIM mice. We did pathway analysis to find altered pathway in muscle tissue. Metabolites from polyamine and beta-alanine pathway were altered.

Effect of ketogenic diet on the plasma and tumor metabolome of melanoma-bearing mice

Project description:Growing evidence supports the use of low-carbohydrate/high-fat ketogenic diets (KDs) together with standard therapies to improve cancer treatment outcomes. However, conflicting data exist regarding the efficacy of KDs in melanoma, the deadliest skin cancer. Here, we show that two different KD formulations effectively reduced tumor growth in immunocompromised mice bearing genetically and metabolically heterogeneous human melanoma xenografts. Furthermore, the KDs exerted a metastasis-reducing effect in an immunocompetent syngeneic melanoma mouse model. Ketone bodies did not directly influence melanoma cell proliferation; therefore, we performed an in-depth metabolomics analysis using the MxP® Quant 500 kit combined with a acylcarnitine assay (biocrates life sciences ag)to elucidate potential anti-tumor mechanisms in vivo. Targeted analysis of plasma and tumor metabolomes revealed distinct changes in amino acid metabolism induced by the KDs. Moreover, the KDs increased sphingomyelin synthesis and hydroxylation of certain lipids. Thus, KDs simultaneously affect multiple metabolic pathways to create an unfavorable environment for melanoma cell proliferation, supporting their potential as a complementary nutritional approach to melanoma therapy.

Extraction of Entamoeba histolytica membrane proteins using three extraction methods

Project description:Entamoeba histolytica membrane proteins are important players in the parasite’s pathogenicity. However, most of the proteins have not been identified. This study reports the membrane proteins extracted using three extractions methods: two commercial kits (ProteoExtract® from Calbiochem and ProteoPrep® from Sigma), and a conventional laboratory method. The resulting membrane fractions (MF) and cytosolic fractions (CF)were analysed using LC-ESI-MS/MS. The proteins identified in at least two out of three biological replicates revealed a total of 490, 492, and 587 MF proteins extracted using the ProteoExtract® kit, ProteoPrep® kit and conventional method, respectively. Meanwhile, 487, 611 and 343 proteins were identified in the CF extracted using the ProteoExtract® kit, ProteoPrep® kit and conventional method, respectively. Analysis of the identified MF and CF proteins extracted by the respective extraction kits suggests that the ProteoPrep® extraction kit was the most selective in separating MF and CF among the three extraction methods.

Pulldown of P180/RRBP1 interacting protein

Project description:P180/RRBP1 is an integral ER-resident protein known to be involved in mRNA binding and translation at the ER. We found that P180/RBBP1 is enriched in neuronal axons. To acquire more knowledge about its interactome and confirmation that P180/RRBP1 is interacting with ribosomes, we performed an unbiased screen of P180 interacting proteins by performing streptavidin pulldowns with GFP-biotin (GFPbio/AviTag)-tagged P180 and subsequent mass spectrometry analysis using HEK293T lysates and adult rat brain extracts. Co-transfection with a BirA construct ensures biotinylation ofP180 and allows subsequent streptavidin pulldown. A GFP-biotin/AviTag only construct was used as control conditionfor specificity.
