Project description:This study is a collaboration with Phil Spence and Jean Langhorne at NIMR, Mill Hill where Phil is studying the differences in aetiology of rodent malaria when mice are infected by mosquito bite versus intraperitoneal injection of blood stage parasites. The aim is to determining transcriptomic differences using RNA-Seq between parasites transmitted by mosquito and those transmitted by injection. This data is part of a pre-publication release. For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/Protocol: Mice were bled out at 6 days post-infection, and RNA was extracted from purified parasite populations using Trizol reagent and DNase treated. Poly A+ mRNA was purified from total RNA using oligo dT dyna bead selection and libraries were created using a modified RNA-seq protocol, where RNA was fragmented using Covaris AFA sonication instead of metal ions. The samples were sequenced on an Illumina HiSeq 2000.

Project description:Plasmoidium chabuadi AS infections initiated by mosquito transmitted infections give rise to an attenuated blood-stage infection when compared to serially blood passaged parasites. Furthermore, Plasmoidium chabuadi CB infections have an increased virulence as compared with Plasmoidium chabuadi AS. We compared the immune response in the spleens induced by these two types of parasites and two types of infection routes in order to determine what may cause this attenuation. Spleens were taken at several timepoints during the blood-stage infection and RNA-seq analysis was performed.

Project description:The glioblastoma is the most malignant form of brain cancer. Glioblastoma cells use multiple ways of communication with the tumor microenvironment in order to tune it for their own benefit. Among these, extracellular vesicles have emerged as a focus of study in the last few years. Extracellular vesicles contain soluble proteins, DNA, mRNA and non-coding RNAs with which they can modulate the phenotypes of recipient cells. In this review we summarize recent findings on the extracellular vesicles-mediated bilateral communication established between glioblastoma cells and their tumor microenvironment, and the impact of this dialogue for tumor progression and recurrence.

Project description:The classical quorum-sensing (QS) model is based on the assumption that diffusible signaling molecules accumulate in the culture medium until they reach a critical concentration upon which expression of target genes is triggered. Here we demonstrate that the hydrophobic signal N-hexadecanoyl-L-homoserine lactone, which is produced by Paracoccus sp., is released from cells by the aid of membrane vesicles (MVs). Packed into MVs, the signal is not only solubilized in an aqueous environment but is also delivered with varying propensities to different bacteria. We propose a novel MV-based mechanism for binary trafficking of hydrophobic signal molecules, which may be particularly relevant for bacteria that live in open aqueous environments.

Project description:Extracellular vesicles and their contents are gaining recognition as important mediators of intercellular communication through the transfer of bioactive molecules such as non-coding RNA. We evaluated the contribution of EV miRNA in intercellular communication between hepatocellular cancer cells and hepatic stellate cells. EV-based miRNA was comprehensively assessed in both cell types individually. Using co-cultures of both HCC and HSC cells in transwell and 3D spheroids culture, we established EV-based miR-126-3p as a potential EV-based miRNA mediator of HSC to HCC communication through which tumor cell migration, invasion and three-dimensional growth in spheroids could be influenced. Manipulation of miR-126-3p by enforced expression or inhibition using pre-miR-126-3p or antimiR-126-3p respectively did not alter cell viability, proliferation, or sensitivity to either sorafenib or regorafenib. In contrast, migration was decreased in HepG2 cells with enforced expression of miR-126-3p. Knockdown of miR-126-3p in HepG2 resulted in a significant increase in ADAM9 expression and in LX-2 cells increased collagen accumulation and the compactness of spheroids but did not alter the number or size of spheroids. The restoration of miR-126 in 3D-co-culture of HepG2 and LX2 cells with precursor showed significant alleviated expression of ADAM9 and VEGF, While the silencing of miR-126 was elevated the expression of ADAM9 and VEGF. These studies implicate miR-126-3p in functional effects on migration, invasion, and spheroid growth in HCC cells in the presence of HSC, and thus demonstrate the presence of functional EV RNA based intercellular signaling between HSC and HCC cells that may be relevant to tumor cell behavior.

Project description:Objectives: MicroRNA (miRNA) can be released to the extracellular medium and participates in neuronal communication. We investigate the mechanisms of miRNA exocytosis by vesicle fusion as a neuromodulator in a manner that are disparate from silencing gene expression. Methods: Small RNA sequencing data of large dense-core vesicle were generated by next-generation sequencing (NGS) in triplicate using Illumina Hiseq 2500. Results: Large dense-core vesicles contain a variety of known and novel miRNAs inside including miR-375. Conclusion: miRNAs can be novel neuromodulators, which are stored in LDCVs and released by vesicle fusion by SNARE assembly and synaptotagmin-1

Project description:The interplay between liver and bone metabolism remains largely uncharacterized. Here, we uncover a mechanism of liver-bone crosstalk regulated by hepatocyte SIRT2. We demonstrate that hepatocyte SIRT2 expression is increased in aged mice and elderly humans. Liver-specific SIRT2 deficiency inhibits osteoclastogenesis and alleviates bone loss in mouse models of osteoporosis. We identify leucine-rich alpha-2-glycoprotein 1 (LRG1) as a functional cargo in hepatocyte-derived small extracellular vesicles (sEVs). In SIRT2-deficient hepatocytes, LRG1 levels in hepatocyte-derived sEVs are upregulated, leading to increased transfer of LRG1-containing sEVs to bone marrow-derived monocytes (BMDMs) and in turn to inhibition of osteoclast differentiation via reduced nuclear translocation of NF-κB p65. Treatment with sEVs carrying high levels of LRG1 inhibits osteoclast differentiation in human BMDMs and in mice with osteoporosis, resulting in attenuated bone loss in mice. Furthermore, the plasma level of sEVs carrying LRG1 is positively correlated with bone mineral density in humans. Thus, drugs targeting hepatocyte-osteoclast communication may constitute a promising therapeutic strategy for primary osteoporosis
