Project description:Hydroxylated polychlorinated biphenyls are the metabolites produced from polychlorinated biphenyls (PCBs) by drug-metabolizing enzyme cytochrome P450 1A1. These compounds are bound to transthyretin, a major plasma thyroid hormone-binding protein in amphibian tadpoles. The compounds-transthyretin complexes are transferred into the brain across the blood brain barrier in mammals. Thus these compounds are suspected to disrupt neural development in brain. We studied about the effects of hydroxylated PCBs on the thyroid system in brain using metamorphosing tadpoles of African clawed toad, Xenopus laevis. The metamorphosis assay revealed that these compounds had inhibitory effects on the thyroid hormone-induced metamorphosis. This in vivo assay was a powerful tool to detect thyroid-disrupting activities, because we were not able to detect the inhibitory effects of these compounds using thyroid hormone-responsive reporter gene assay in a cultured Xenopus cell line. A genome-wide gene expression analysis in brain following short-term exposure to these compounds demonstrated that the delay of metamorphosis and the morphological thyroid-disrupting changes could be caused partially by disruption of the thyroid hormone-induced gene expression by hydroxylated PCBs. Furthermore, we associated functional ontology terms with the transcripts whose expression were altered by thyroid hormone alone, or thyroid hormone and hydroxylated PCBs. We suggested that these approachs using a technique of bioinformatics revealed molecular mechanism of thyroid-disrupting activities in vivo.

Thyroid hormones induce amphibian metamorphosis and alter a lot of thyroid hormone-responsive gene expression. We studied about the effects of hydroxylated PCBs on TH-induced gene expression. Premetamorphic tadpoles were treated with 500 nM hydroxylated PCBs in the presence of 1 nM thyroid hormone for 4 days. After exposure period total RNA was extracted from brain. Study included at least three replicate of each treatment.

Project description:Thyroid hormone improves left ventricular remodeling and cardiac performance after myocardial infarction (MI), but the molecular basis is unknown. This study was designed to detect gene expression changes in left ventricular non-infarcted areas at 4 weeks following myocardial infarction with and without thyroid hormone treatment. The results suggest that altered expression of genes for molecular function and biological process may be involved in the beneficial effects of thyroid hormone treatment following myocardial infarction in rats.

Project description:Animals adapt to environmental conditions by modifying the function of their internal organs, including the brain. To be adaptive, alterations in behavior must be coordinated with the functional state of organs throughout the body. Here we find that thyroid hormone--a regulator of metabolism in many peripheral organs--directly activates cell-type specific transcriptional programs in frontal cortex of adult male mice. These programs are enriched for axon-guidance genes in glutamatergic projection neurons, synaptic regulatory genes in both astrocytes and neurons, and pro-myelination factors in oligodendrocytes, suggesting widespread plasticity of cortical circuits. Indeed, whole-cell electrophysiology revealed that thyroid hormone alters excitatory and inhibitory synaptic transmission, an effect that requires thyroid hormone-induced gene regulatory programs in presynaptic neurons. Furthermore, thyroid hormone action in frontal cortex regulates innate exploratory behaviors and causally promotes exploratory decision-making. Thus, thyroid hormone acts directly on cerebral cortex in males to coordinate exploratory behaviors with whole-body metabolic state.

Project description:Although it is well known that the thyroid hormone (T3) is an important positive regulator of cardiac function over a short-term and that it also promotes deleterious effects over a long-term, the molecular mechanisms for such effects are not well understood yet. Since most alterations in cardiac function are associated to changes on sarcomeric machinery, the present work was undertaken in order to find novel sarcomeric hot spots driven by T3 in the heart. We analysed gene expression changes in heart of rats submitted to experimental hyperthyroidism through microarray analysis. The timeline analysis included euthyroid group as control, 12, 24 hours and 7 days treatment groups.

Project description:Juvenile hormone (JH) and JH agonists have been reported to induce male offspring production in various daphnids including Daphnia magna. We have recently established a short-term in vivo screening method to detect chemicals having JH activity, induction of male offspring, using adult D. magna. Diofenolan has been developed to be a JH agonist in insects, but no male induction in daphniids has been studied. In this study, we found the male offspring induction by diofenolan with a short-term in vivo screening method, and the most potent male offspring production activity among JH analogs studied using the OECD TG211 Annex 7. Transcriptome analysis of diofenolan-exposed D. magna showed up-regulation of JH-responsive genes. These results clearly demonstrated that diofenolan has JH activities in D. magna using the short-term in vivo screening assay, OECD TG211 Annex 7 and microarray analysis.
