Project description:The human testis undergoes dramatic developmental and structural changes during puberty, including proliferation and maturation of niche/somatic cells, and the onset of spermatogenesis. Here, we profiled and analyzed single-cell transcriptomes of ~10,000 testicular cells from four boys spanning puberty, and compared to infants and adults. During puberty, undifferentiated spermatogonia first expand and then differentiate, prior to gametogenesis. Notably, we identify a common pre-pubertal progenitor for Leydig and myoid cells, and reveal candidate factors/pathways for pubertal differentiation. Furthermore, pre-pubertal Sertoli cells form two states that differ in mitochondrial/metabolic transcription, which converge to a single mature population during puberty. Roles for testosterone in Sertoli cell maturation, antimicrobial peptide secretion and spermatogonial differentiation are revealed through analyses of testosterone-suppressed transgender female testis and via in vitro seminiferous tubule culturing. Overall, our transcriptional atlas of the developing human testis provides major insights into developmental changes and key factors/pathways that accompany male puberty.

Project description:The successful proliferation and differentiation of testicular cells are vital for the survival, continuity and preservation of species. However, a comprehensive molecular orchestration underlying the spermatogenic process in teleost testis development throughout the annual cycle remains elusive. In this study, the testicular cells derived from adult black rockfish at distinct stages—Regressed, Regenerating, and Differentiating were dissected via single cell transcriptome sequencing. A continuous developmental trajectory of spermatogenic cells, from spermatogonia to spermatids, was delineated, elucidating the molecular events involved in the spermatogonia development. Subsequently, the dynamic regulation of gene expressions associated with spermatogonia proliferation and differentiation was observed across spermatogonia subgroups and various developmental stages. A bioenergetic transition from glycolysis to mitochondrial respiration of spermatogonia during annual developmental cycle was demonstrated, and a deeper level of heterogeneity and molecular characteristics was revealed by re-clustering analysis. The developmental trajectory of Sertoli cells in annual reproductive cycle was examined, and the divergence of Leydig cells and macrophage were explored. Additionally, the interaction network between testicular micro-environment somatic cells and spermatogenic cell were established. Overall, our study provides detailed information on both germ and somatic cells within teleost testis during annual reproductive cycle, along with insights into the molecular regulation of spermatogonia dynamics, which lay the foundation for spermatogenesis regulation and germplasm preservation of endangered species.

Project description:We have employed whole genome microarray expression profiling as a discovery platform to identify the effect of plin1 on gene expression during testis development. Testis of day 21, day 40, and day 60 in plin1-null mice and wild type mice were analyzed. Expression of a list genes (including cyp51, prm1, tnp2) from this signature was quantified in the same RNA samples by real-time PCR, confirming the influence of plin1 on testis development. plin1 knockout alteration gene expression in mice testis was measured at different development phases: day 21 meiosis phase, day 40 puberty phase, and day 60 mature phase. There independent experiments were performed at each time (d21, d40, d60).

Project description:When puberty starts before males reach harvest size, animal welfare and sustainability issues occur in Atlantic salmon (Salmo salar) aquaculture. Hallmarks of male puberty are an increased proliferation activity in the testis and elevated androgen production. Examining transcriptional changes in salmon testis during the transition from immature to maturing testes may help understanding the regulation of puberty, potentially leading to procedures to modulate its start. Since differences in body weight influence, via unknown mechanisms, the chances for entering puberty, we used two feed rations to create body weight differences. Maturing testes were characterized by an elevated proliferation activity of Sertoli cells and of single undifferentiated spermatogonia. Pituitary gene expression data suggest increased Gnrh receptor and gonadotropin gene expression, potentially responsible for the elevated circulating androgen levels in maturing fish. Transcriptional changes in maturing testes included a broad variety of signaling systems (e.g. Tgfβ, Wnt, insulin/Igf, nuclear receptors), but also, activation of metabolic pathways such as anaerobic metabolism and protection against ROS. Feed restriction lowered the incidence of puberty. In males maturing despite feed restriction, plasma androgen levels were higher than in maturing fish receiving the full ration. A group of 449 genes that was up-regulated in maturing fully fed fish, was up-regulated more prominently in testis from fish maturing under caloric restriction. Moreover, 421 genes were specifically up-regulated in testes from fish maturing under caloric restriction, including carbon metabolism genes, a pathway relevant for nucleotide biosynthesis and for placing epigenetic marks. Undifferentiated spermatogonia and Sertoli cell populations increased at the beginning of puberty, which was associated with the up-regulation of metabolic pathways (e.g. anaerobic and ROS pathways) known from other stem cell systems. The higher androgen levels in males maturing under caloric restriction may be responsible for the stronger up-regulation of a common set of (449) maturation-associated genes, and the specific up-regulation of another set of (421) genes. The latter opened regulatory and/or metabolic options for initiating puberty despite feed restriction. As a means to reduce the incidence of male puberty in salmon, however, caloric restriction seems unsuitable.
