Several studies have reported sex-related differences in the structure and function of human brains. Oligodendrocyte precursor cells (OPCs) regulate the neuronal system in various ways and play crucial roles in brain homeostasis besides their well-known role as a reservoir for mature oligodendrocytes. Here, we revealed transcriptomic differences in OPCs isolated from male and female neonatal rat brains.

Project description:Glial cytoplasmic inclusion is the pathological hallmark of multiple system atrophy commonly observed as alpha-synulcien-positive aggregates within oligodendrocytes. We elucidated that preformed alpha-synulcien fibrils triggers multimerization and upsurge of endogenous alpha-synulcien in oligodendrocyte precursor cells which is attributable to insufficient autophagic proteolysis. To assess the functional influence of alpha-synuclein fibrils application on oligodendrocyte precursor cells, RNA-seq analysis was performed. The data revealed that alpha-synulcien fibrils interfered with numerous protein expressions associated with neuronal support and myelination.

Investigation of Oligodendrocyte Precursor Cell Differentiation by Quantitative Proteomics

Project description:Oligodendrocytes, the myelinating cells of the central nervous system, are essential for correct brain function. They originate from oligodendrocyte precursor cells (OPCs) through a differentiation process which is only incompletely understood and impaired in a variety of demyelinating diseases. Better knowledge of this differentiation holds the promise to develop novel therapies for these disorders. We investigated the differentiation of rat oligodendrocyte precursor cells to oligodendrocytes in vitro. After confirmation of differentiation by immunohistochemical analysis using cell type-specific marker proteins, we conducted a quantitative proteomics study using tandem mass tags (TMT). We investigated four time points of differentiation covering early, intermediate, and late stages. Data analysis was perfomed using Mascot and Maxquant.

Expression profiles in Chd7 knockdown oligodendrocyte precursor cells

Project description:Oligodendrocyte precursor cells (OPCs) regulate neuronal, glial and vascular systems in diverse ways and display phenotypic heterogeneity beyond their well-known role as a reservoir for mature oligodendrocytes. Here we demonstrated that perivascular OPCs in cerebral cortex were increased coupling with post-stroke angiogenesis in a mouse model of stroke induced by middle cerebral artery occlusion (MCAO). RNA-seq analysis revealed that primary cultured OPCs notably increased the gene expressions of numerous pro-angiogenic factors, while decreased those related to OPC maturation after oxygen glucose deprivation. The data showed that cerebral ischemia shifts the phenotype of OPCs to perivascular subtypes which can promote angiogenesis.

The difference of gene expression in mouse OPCs in normothermic and hypothermic culture

Project description:We have found that the cell yield of oligodendrocyte precursor cells (OPCs) are higher in 31.5 degC than in 37 degC not by suppression of apoptosis but by enhancement of proliferation. Here, we performed transcriptome analysis using Affymetrix GeneChip on the primary cultured oligodendrocyte precursor cells prepared from the brain of ICR mice. Oligodendrocyte precursor cells were prepared from primary mixed cell cultures of embryonic mouse cerebral hemispheres. The cerebral hemispheres from 15-day-old mouse embryos were enzymatically dissociated, seeded and cultured for 5 days in D-MEM containing 10% FBS. The cells were passaged to serum-free basal medium and cultured for further 48 hr at 37 degC and 31.5 degC.
