Gene expression response of wolf epidermal keratinocytes to PolyI:C and canine distemper virus

Project description:The goal of the study was to test whether CBD103 genotype of North American gray wolves impacts the gene expression response to polyI:C or to live canine distemper virus. We established 24 primary cultures of epidermal keratinocytes from skin punches of North American gray wolves, and also generated an immortalized keratinocyte line and a CRISPR/Cas9 edited cell line. We evaluated the gene expression response of cells to either 24 hours challenge with 1 ug/ml polyI:C or to five days challenge with live canine distemper virus (100 TCID50/ml). Every challenged cell culture had a paired null control sample (plated and collected at same time points).

Project description:An anthropogenic chemical contaminant commonly identified in aquatic receiving environments is the non-steroidal anti-inflammatory drug (NSAID), ibuprofen(IBF). While the role of ibuprofen in target organisms is known, there exists a paucity of data on the impact of exposure to non-target wildlife species. In the case of frog species, normal development and environmental fitness involves the actions of the thyroid hormones (THs), particularly at key points in the life cycle. We investigated whether exposure of premetamorphic North American bullfrog (Rana catesbeiana) tadpoles to IBF altered their response to treatment with an exogenous dose of thyroid hormone (T3).

Identification and characterization of amphibian SLC26A5 using RNA-Seq

Project description:We mapped and sequenced the SLC26A5 of the American bullfrog from its inner ear cDNA using RNA-Seq. The frog SLC26A5 cDNA was 2,292 bp long, encoding a polypeptide of 763 amino acid residues, with 40% identity to mammals. After isolating the prestin gene of the frog, we generated a stable cell line transfected with this new coding gene and found it possessing similar electrophysiological features as the hair cells from the frog’s auditory organ. Our experiment demonstrated that the new coding gene could encode a functionally active protein conferring NLC to both frog HCs and the mammalian cell line.
