Expression profile of the fat body from bigmax mutant and wildtype third instar Drosophila melanogaster larvae

Project description:To identify genes differentially expressed in the fatbody of Drosphila melanogaster bigmax mutants, a loss-of-function allele was generated by P-element mobilization. Mutant and wildtype first instar larvae were raised on two different sources of food, control and high-sugar media. When the animals reached the wandering third instar stage, animals were sacrificed and their fat bodies dissected. Total RNA was extracted, labeled fluorescently and hybridized competitively to Agilent's 4x44K Drosophila Gene Expression Microarrays. On each array, three different samples were analyzed: 1. wildtype animals raised on control food, 2. wildtype animals raised on high-sugar food and 3. bigmax mutant animals raised on high-sugar food.

Glucose-dependent Insulinotropic polypeptide receptor-expressing cells in the hypothalamus regulate food intake

Project description:Ambiguity regarding the role of glucose-dependent insulinotropic polypeptide (GIP) in obesity arises from conflicting reports asserting that both GIP receptor (GIPR) agonism and antagonism are effective strategies for inhibiting weight gain. To enable identification and manipulation of Gipr-expressing (Gipr) cells we created GIPR-Cre knock-in mice. As GIPR-agonists have recently been reported to suppress food intake we aimed to identify central mediators of this effect. Gipr cells were identified in the arcuate, dorsomedial, and paraventricular nuclei of the hypothalamus, as confirmed by RNAscope in mouse and human. Single cell RNAseq identified clusters of hypothalamic Gipr cells exhibiting transcriptomic signatures for mural, glial and neuronal cells, the latter expressing somatostatin, but little proopiomelanocortin or agouti-related peptide. Activation of Gq-DREADDs in hypothalamic Gipr cells suppressed food intake in vivo, which was not obviously additive with concomitant GLP1R activation. These data identify hypothalamic GIPR as a target for the regulation of energy balance.

Transcription profiling of Drosophila Roo lines grown on lead containing food to investigate lead toxicity

Project description:75 Diosophilia Roo lines, recominant inbred lines, were raised to adulthood either on conventional diet or diet supplemented with lead acetate. Whole flies were used for RNA extraction. RNA was run on Dros Genome 2 arrays. Each line also genotyped for 88 markers; Control food consisted of standard cornmeal, agar, sugar, yeast, and 250 microM NaAc (Ashburner 1989). Lead-contaminated food consisted of standard food plus 250 microM PbAc (lead exposure at this concentration has been shown to affect locomotion in adults (Hirsch et al. 2003)). Genetic and genomic response to selection for food consumption in Drosophila melanogaster

Project description:Food consumption is critical for animal survival and reproduction. The biomedical and economic consequences of metabolic diseases arising from excessive food intake, however, are a burden for human society. While the role of neuroendocrine feedback loops, food sensing modalities, and physiological state in regulating food intake are increasingly well understood, other genetic mechanisms remain elusive. Here, we applied ten generations of artificial selection for high and low food consumption in replicate populations of Drosophila melanogaster. The phenotypic response to selection was highly asymmetric, with efficient selection and an average realized heritability of 0.15 in the lines selected for high food consumption. To further nominate candidate genes contributing to response to selection for feeding behavior, we evaluated differences in genome wide gene expression between the selection lines using whole-fly RNA sequencing. We identified 1,631 differentially expressed genes in the analysis pooled across sexes, and 1,267 (2,321) differentially expressed genes in females (males).

Project description:C57BL6/J female mice were raised either in standard environment (SE) for 8 weeks after birth or were put in enriched environment (EE) at postnatal 4 weeks up to 8 weeks old. Mice were sacrificed at 8 weeks and spleen were harvested. CD4+ and CD8+ T cells were sorted by FACS and CD4+ or CD8+ T were profiled using a low-input protocol adapted from single-cell RNAseq.
