Project description:Changes in gene expression during berry development during a grape growing season were analysed. The effect on gene expression of different viticultural practises during grape berry development was investigated in this study by comparing two pruning methods (spur versus machine). Grape berries were collected and pooled on a weekly basis to obtain a developmental series comprised of 17 developmental stages from flowering until harvest across the grape growing season for both spur and machine pruned vines. Project description:Human aortic endothelial cells were grown in culture until confluent. In three experiments using cells derived from three separate donors confluent cultures were incubated for 6 h with contol medium, or medium containing either extracts of oligomeric procyanidins from cranberry juice or red wine, or a procyanidin-rich grape seed extract. At the end of the 6 h treatment period conditioned media samples were retained for immunoassay of secreted peptides and proteins, and RNA was extracted for microarray analysis. Experiment Overall Design: Each experiment used cells from one donor. Treatment conditions were: control medium, cranberry extract (CRE), grape seed extract (GSE), and red wine extract (RWE).
Project description:We analysed the molecular effects on gene modulation exerted by aglianico grape seed extract (AGS) in mesothelioma. Previous results allowed to demonstrate that AGS polar extracts determined apoptosis in three different mesothelioma cell lines including one resistant to conventional chemotherapeutic treatment. In addition this treatment affected tumorigenic and invasive properties of these cells We used microarrays to identify molecular pathway deregulated by AGS treatment and involved in apoptosis induction.
