Project description:Nematodes such as Steinernema carpocapsae are used as organic pesticides because of their ability to prey on live insects. They do so thanks to their symbiotic bacteria, that they release within the hemocoel of insects. In this study we wanted to study how Spodoptera frugiperda, a Lepidoptera pest of crops becoming invasive around the world, defend themselves against the nematodes. We infested S. frugiperda larvae with nematodes and dissected three tissues: the midgut, the fat body and the hemocytes at two time-points: 8 h and 15 h after infestation. We performed single-end RNA-seq on these samples to study the tissue specificity of S. fru response, as well as its dynamic.

Project description:Pesticides are widely used to protect growth of crops and prevent the spread of diseases. However, more and more pest insects have developed resistance to chemical pesticides along with the long-term application of the pesticides. It is very important to explore the molecular mechanisms of insecticide resistance not only for reversing the resistance in insects, but also for finding out new function targets of the insecticides. Recently, the next-generation sequence technique has become an effective tool to screen resistance genes and has developed transcriptome profiles of various species. However, a comprehensive database to collect these transcriptome data remains poorly developed. In this study, we constructed a database for insect resistance called IRdb, which contains gene count data from various insect species analyzed by a unified process. In addition to the gene data, IRdb also contains 430 unique resistance proteins (experimentally verified proteins manually extracted from literature). Users can discriminate the resistance proteins by submitting fasta sequence of proteins of interest, which can provide clues to detect resistance proteins. The application of resistance protein part in IRdb indicates the accuracy of prediction of IRdb by extracting CTD features and employing random forest. The database IRdb online web server (http://120.27.24.199:20609/) was provided for users to download the transcriptome and protein data for resistance of insects to insecticides and to predict potential resistance proteins.

Project description:N-glycosylation is one of the most abundant and conserved protein modifications in eukaryotes. This modification serves various important functions, such as protein folding and cellular attachment, but also modulation of a protein’s function. Recently, it has been shown that N-glycosylation of proteins plays a vital role in insect development and survival, which makes it an interesting target for pest control. Despite the importance of protein N-glycosylation in insects, not much is known about insect N-glycoproteomes. Here, we report on the N-glycoproteomes of three major pest insects spanning different insect orders; Drosophila melanogaster (Diptera), Tribolium castaneum (Coleoptera) and Acyrthosiphon pisum (Hemiptera). The number of identified N-glycosylation sites ranged from 889 in T. castaneum, to 941 in D. melanogaster and 1,338 in A. pisum. Comparison between the different insect species revealed both conserved and species-specific glycoproteins. The functionality of the insect glycoproteins together with the conservation of the N-glycosites throughout evolution are discussed. This information can help in the elaboration of novel pest insect control strategies based on interference in insect glycosylation.

Project description:The role of plant-derived food sources in plant-herbivore-carnivore interactions is an essential topic in basic ecology, and it is directly linked to the applied discipline of biological pest control. Honeydew, which phloem-feeding hemipteran insects excrete, constitutes a rich source of carbohydrates and proteins for beneficial insects in ecosystems. However, the quality of this plant-derived food source depends on its composition. Here we demonstrated that plant proteins in honeydew increase the fitness of a parasitic wasp (member of the third-trophic level).

Project description:N6-methyladenosine RNA (m6A) is the most abundant modification of messenger RNA (mRNA), which has been demonstrated in regulating various post-transcriptional processes and playing key roles in sex determination, neuronal functions, and embryonic development in Drosophila and mice. However, the methylation profile and relevant functions in other insects remain largely unknown. Here, we analyzed transcriptome-wide profile of m6A modification in the embryonic development of a destructive agricultural pest Spodoptera frugiperda. Our study showed that differentially expressed m6A peak-modified genes were enriched in cell and organ development between the 2 h and 24 embryos. Our study reveals that RNA m6A epigenetic regulation in the embryonic development of S. frugiperda, and provides new insight for the embryonic development of insects.

These inhibitors arrest proteolytic digestion by binding to the proteases in the gut. However, many pest insects have coevolved mechanisms to counteract these effects. While the nature of these counter-defensive strategies has been well described for several insect pests, our understanding of how these responses are triggered and regulated remains limited. In this study we investigated the initiation of this adaptive response in the economically damaging migratory locust (Locusta migratoria), via microarray analysis of gut tissues following the dietary uptake of PIs. For this purpose, the total number of ESTs available for locusts was narrowed down to transcripts that were found expressed in gut and/or brain tissue. The remaining subset was used for the construction of an optimized microarray platform.
