Project description:Upon weightlessness and microgravity, deleterious effects on the neurosensory and neurovestibular systems, haematological changes, and deconditioning of musculoskeletal, cardiovascular and cardiopulmonary functions have been reported. In particular, loss of muscle mass and strength are triggered by weightlessness in humans during space flights, what is similarly observed as a result of physical inactivity conditions and ageing on Earth. However, skeletal muscle tissue is of paramount importance for health maintenance (e.g. being essential to locomotion, heat production, and metabolism). To better prevent or eventually treat microgravity-induced muscle atrophy, its underlying mechanisms have first to be characterized in details. Using cutting-edge quantitative proteomics, the aim of the present study was therefore to get an in depth view of the molecular regulations triggered by space conditions in skeletal muscles of mice during the 30-day flight of the BION-M1 biosatellite. As muscles differing in their fiber type composition appear to respond differently to microgravity (see above), we characterized here the differential response of the soleus, extensor digitorum longus and vastus lateralis muscles.

Project description:Bone loss and immune dysregulation are among the main adverse outcomes of spaceflight challenging astronaut’s health and safety. However, consequences on B cell development and responses are still under-investigated. Up to now, most studies addressing these questions were performed using an amphibian species. Consequently, we used advanced proteomics analysis of femur bone and marrow of mice flown for one month on board the BION-M1 biosatellite, to determine whether extreme conditions encountered during a real spaceflight affect B cell development in mice and to examine reversibility of the effects upon return to Earth. Our data revealed that adverse effects on B lymphopoiesis were more marked one week after landing and that this phenomenon was associated with a 41% reduction of B cells in the spleen. Thus, the effects of spaceflight persisted during at least one week after landing. These reductions may contribute to explain increased susceptibility to infection even if we confirmed that animals were able to mount a humoral immune response.

Project description:Ageing is characterized by the progressive lowering of the acute innate immune response and the progressive up-regulation of low-grade inflammation (i.e. inflammaging). At the cellular level, telomeres are considered as a mark of ageing as their length is progressively shortened. However, the links between telomeres and innate immune ageing remain undefined. Using a proteomic approach conducted on laboratory mice, we tested whether the experimental triggering of an acute innate response is deleterious for telomere maintenance, and whether this is dependent of age. To do so, we challenged young and old mice using bacterial lipopolysaccharide (LPS) and measured individuals’ end-points of telomere length and proteomic profiles in spleen tissue.

Project description:Muscle atrophy is one of the main deleterious consequences of ageing and physical inactivity. Although basic knowledge regarding the underlying mechanisms of muscle atrophy is continuously growing, there are still no efficient therapeutic strategies for its prevention and treatment. Hibernating bears exhibit a strong and unique ability to preserve muscle mass in conditions where muscle atrophy is observed in humans. However, underlying mechanisms have not been understood yet. To fill this gap, the aim of this study was to characterize changes in the bear muscle proteome during hibernation versus the active period. Muscle biopsies were obtained from Ursus arctos bears.

Project description:Protein expression is regulated by production and degradation of mRNAs and proteins, but their specific relationships remain unknown. We combine measurements of protein production and degradation and mRNA dynamics to build a quantitative genomic model of the differential regulation of gene expression in LPS stimulated mouse dendritic cells. Changes in mRNA abundance play a dominant role in determining most dynamic fold changes in protein levels. Conversely, the preexisting proteome of proteins performing basic cellular functions is remodeled primarily through changes in protein production or degradation, accounting for over half of the absolute change in protein molecules in the cell. Thus, the proteome is regulated by transcriptional induction of novel cellular functions and remodeling of preexisting functions through the protein life cycle. Mouse primary dendritic cells were treated with LPS or mock stimulus and profiled over a 12-hour time course. Cells were grown in M-labeled SILAC media, which was replaced with H-labeled SILAC media at time 0. Aliquots were taken at 0, 0.5, 1, 2, 3, 4, 5, 6, 9, and 12 hours post-stimulation and added to equal volumes of a master mix of unlabeled (L) cells for the purpose of normalization. RNA-Seq was performed at 0, 1, 2, 4, 6, 9, and 12 hours post-stimulation.

Project description:Animal longevity widely differs across species, and even individuals from the same species may exhibit different rates of ageing. In different species, the rate at which individuals actually age is related to the level of their social interactions, but this was still not known in ants. In a given colony, ant individuals are close genetic relatives, exhibit very different behaviours and a contrasted lifespan according to the Caste. Such characteristics constitute main advantages to study relationships between sociality and ageing. Therefore, the aim of this study was to characterize differences in the proteome of Lasius niger queens versus that of domestic and foraging workers. Proteomic data were put in relation with the behaviour of individuals from the three Castes. Hence, it was found that sociality correlates with ant longevity, with i) social immunity enabling the queen to mainly invest in soma protection, and ii) marked exposition of workers to the environment and nutrients inducing metabolic pathways that reduce their lifespan.

Project description:Grey mouse lemurs (Microcebus murinus) are a primate species, which exhibits strong physiological seasonality in response to environmental energetic constraint. They notably accumulate large amounts of lipid reserves during early winter, which are thereafter mobilized during late winter, when food availability is very low. In addition, they develop glucose intolerance in late winter only. To decipher how the hepatic mechanisms may support such metabolic flexibility, we analyzed the liver proteome of captive lemurs, which seasonal regulations of metabolism and reproduction are comparable to their wild counterparts, during the phases of either constitution or use of fat reserves.

Project description:In social insects, workers perform distinct tasks according to the caste they belong to, and workers from different castes differ in their age (nest workers are usually younger than foragers are). The caste shift thus seems inseparable from age, preventing from deciphering the role of labour division and age in regulating individual physiology and ageing rates. We set up an experimental protocol separating age and caste effects by defining four groups of black garden ant (Lasius niger) workers: young foragers (Y.F), old foragers (O.F), young nest workers (Y.NW) and old nest workers (O.NW). Proteomics highlighted differences between individuals according to their age, whereas metabolomics revealed caste-related differences. Our study highlighted that age and caste influence specifically different aspects of the physiology of ant workers.
