Mass spectrometry analysis of unconventional kinetoplastid kinetochore proteins KKT1, KKT2, and KKT6

ABSTRACT: The kinetochore is the macromolecular protein machine that drives chromosome segregation in eukaryotes. Here we performed chemical crosslinking mass spectrometry of the Trypanosoma brucei KKT2 DPB/KKT1C complex and KKT1/KKT6 complex. We also performed immunoprecipitation and mass spectrometry of KKT1 fragments (KKT1N and KKT1C) that were ectopically expressed and purified from trypanosomes.

Project description:We performed chemical crosslinking mass spectrometry of native protein complexes purified from Trypanosoma brucei procyclic cells to obtain protein-protein interaction information for the chromosomal passenger complex (CPC) and kinetochore complex. Crosslinks among CPC subunits were detected using YFP-AUK1, while those among KKT8 complex subunits were detected in the YFP-KKIP1 sample. We also performed immunoprecipitation and mass spectrometry of CPC subunits (full-length proteins and fragments) to identify co-purifying proteins.

Project description:KKT23 is a kinetoplastid kinetochore protein that has structural similarity to the GCN5 acetyltransferase domain. Using a kinetoplastid parasite Trypanosoma brucei, here we performed immunoprecipitation and mass spectrometry of GFP-tagged KKT23 N-terminal fragment (2–150) to identify co-purifying proteins, and immunoprecipitation and mass spectrometry of H2B-GFP after KKT23 depletion to examine acetylation states of histones. We also performed in vitro acetylation assays on recombinant trypanosome nucleosomes using recombinant KKT23 protein, followed by mass spectrometry.

BioID of Plasmodium falciparum rhophtry-neck associated protein (PfAIP) using BirA* fusion-protein

Project description:Apicomplexan parasites contain rhoptries, which are specialized secretory organelles that coordinate host cell invasion. During the process of invasion, rhoptries secrete their contents to facilitate interaction with, and entry into, the host cell. Here we report the crystal structure of the rhoptry protein Armadillo Repeats-Only (ARO) from the human malaria parasite, Plasmodium falciparum (PfARO). The structure of PfARO is comprised of five tandem Armadillo-like (ARM) repeats, with adjacent ARM repeats stacked in a head-to-tail orientation resulting in PfARO adopting an elongated curved shape. Interestingly, the concave face of PfARO contains two distinct patches of highly conserved residues that appear to play an important role in protein-protein interaction. We functionally characterized the P. falciparum homologue of ARO interacting protein (PfAIP) and demonstrate that it localizes to the rhoptries. We show that conditional mislocalization of PfAIP leads to deficient red blood cell invasion. Guided by the structure, we identified mutations of PfARO that lead to mislocalization of PfAIP. Using proximity-based biotinylation we probe into PfAIP interacting proteins.
