ITRAQ based proteomic analysis to reveal the mechanism of tigecycline resistance in Acinetobacter baumannii

ABSTRACT: The antibiotic resistance of A. baumannii has been increasing in recent years. There are still many questions unclear concerning the mechanism of tigecycline resistance in A. baumannii. iTRAQ based proteomic analysis were used to reveal the mechanism of tigecycline resistance in Acinetobacter baumannii.

Project description:Using Nanopore sequencing, our study has revealed a close correlation between genomic methylation levels and antibiotic resistance rates in Acinetobacter Baumannii. Specifically, the combined genome-wide DNA methylome and transcriptome analysis revealed the first epigenetic-based antibiotic-resistance mechanism in A. baumannii. Our findings suggest that the precise location of methylation sites along the chromosome could provide new diagnostic markers and drug targets to improve the management of multidrug-resistant A. baumannii infections.

Proteomic analysis of Masson pine with high resistance to pine wood nematode

Project description:Pine wilt disease is a worldwide dangerous pine disease. We used Masson pine (Pinus massoniana) clones, selected through traditional breeding and testing for 20 years, with high resistance to study the molecular mechanism of resistance to pine wood nematode (PWN, Bursaphelenchus xylophilus). A total of 3491 proteins were identified from seedling tissue, among which 2783 proteins contained quantitative information. Total 42 proteins were up-regulated and 96 proteins were down-regulated in resistant lines. Of them, function enrichment analysis found that significant differences in proteins with pectin esterase activity or peroxidase activity. Proteins participating in salicylic acid metabolism, antioxidant stress reaction, polysaccharide degradation, glucose acid ester sheath lipid biosynthesis, sugar glycosaminoglycans degradation pathway also changed significantly. PRM results showed that pectin acetyl esterase, carbonic anhydrase, peroxidase and chitinase were significantly down-regulated, while aspartic protease was significantly up-regulated, which was consistent with proteomic data.These results suggested that Masson pine could degrade nematode-related proteins by increasing protease to inhibit their infestation, and enhance the resistance of Masson pine to PWN by down-regulating the carbon metabolism to limit available carbon to PWN or to be involved in cell wall components or tissue softening. Most downregulated proteins seem to take back seats prior to pathogen attacks. The highly resistant Masson pine, very likely, has evolved multiple pathways, both the passive and active, to defense against PWN infestation.

Transcriptional response of Acinetobacter baumannii to hydrogen peroxide and role of mumR and oxyR in regulating this response

Project description:Acinetobacter baumannii is a Gram-negative opportunistic pathogen that causes multiple infections, including pneumonia, bacteremia, and wound infections. Due to multiple intrinsic and acquired drug-resistance mechanisms, A. baumannii isolates are commonly multi-drug resistant and infections are notoriously difficult to treat. Therefore, it is important to identify mechanisms used by A. baumannii to survive stresses encountered during infection as a means of identifying new drug targets. In this study, we determined the transcriptional response of A. baumannii to hydrogen peroxide stress using RNASequencing. Upon exposure to hydrogen peroxide, A. baumannii differentially transcribes several hundred genes. In this study, we also determined the transcriptional profile of A. baumannii strains with the transcriptional regulators mumR or oxyR genetically inactivated and identified transcriptional differences between these strains and wild-type A. baumannii in response to hydrogen peroxide stress. In doing this, the function of A. baumannii OxyR in hydrogen peroxide stress resistance and regulation of genes required for hydrogen peroxide detoxification was defined. Moreover, the contribution of the uncharacterized regulator MumR to hydrogen peroxide stress resistance was also explored. This work reveals the transcriptome of an important human pathogen in the presence of hydrogen peroxide stress.

Colistin-resistant, lipopolysaccharide-deficient Acinetobacter baumannii responds to lipopolysaccharide loss through increased expression of genes involved in the synthesis and transport of lipoproteins, phospholipids and poly-beta-1,6-N-acetylglucosamine

Project description:We recently demonstrated that colistin resistance in Acinetobacter baumannii can result from mutational inactivation of genes essential for lipid A biosynthesis. Consequently, strains harboring these mutations are unable to produce the major Gram negative bacterial surface component, lipopolysaccharide (LPS). To understand how A. baumannii compensates for the lack of LPS, we compared the transcriptional profile of the A. baumannii type strain ATCC19606, to that of an isogenic, LPS-deficient, lpxA mutant strain. Analysis of the expression profiles indicated that the LPS-deficient strain showed increased expression of many genes involved in cell envelope and membrane biogenesis. In particular, up-regulated genes included those involved in the Lol lipoprotein transport system and the Mla-retrograde phospholipid transport system. In addition, genes involved in the synthesis and transport of poly-beta-1,6-N-acetylglucosamine (PNAG) were also up-regulated and a corresponding increase in PNAG production was observed. The LPS-deficient strain also exhibited reduced expression of genes predicted to encode the fimbrial subunit FimA and a type VI secretion system (T6SS). The reduced expression of genes involved in T6SS correlated with the detection of the T6SS-effector protein, AssC, in culture supernatants of the A. baumannii wild-type strain, but not in the LPS-deficient strain. Taken together, these data show that, in response to total LPS loss, A. baumannii alters the expression of critical transport and biosynthesis systems associated with modulating the composition and structure of the bacterial surface.
