Host biomarker discovery for ectoparasite resistance in cattle

ABSTRACT: This project aimed to discover the protein-based biomarkers for tick resistance in cattle using cattle serum samples. The cattle were phenotyped into two groups, tick-resistant and susceptible after an artificial tick challenge. Mean tick scores were used to categorise cattle. The SWATH analysis was sued to measure the relative abundance of proteins in skin samples of the two groups at different time points.

Project description:Previous studies have applied genomics and transcriptomics to identify the immune and genetic markers as key indicator traits for cattle tick susceptibility/resistance, however, results differed between breeds, and host proteomics and metabolomics were not considered. We used serum samples from Santa Gertrudis cattle phenotypically divided into groups as tick-resistant (TR) and -susceptible (TS) to conduct differential abundance analyses of protein profiles between the two groups. The serum proteins were digested into peptides followed by identification and quantification by sequential window acquisition of all instances of theoretical fragment ion mass spectrometry (SWATH-MS).

Transcription profiling of Bos taurus skin of tick naive and tick-infested Holstein-Friesian and Brahman breeds

Project description:This experiment was undertaken to document changes in gene expression in the skin of tick-resistant Brahman (Bos indicus) and tick-susceptible Holstein-Friesian (Bos taurus) cattle prior to, and following, infestation with the cattle tick Rhipicephalus (Boophilus) microplus Experiment Overall Design: RNA was extracted from skin samples of tick-naÃ¯ve cattle (animals with no previous R.microplus exposure) and tick-infested cattle after a period of successive, heavy infestations with R. microplus. Skin samples taken from tick-infested animals were taken at sites where tick larvae (approximately 24 h old) were attached to the skin sample. Skin samples were of 8 mm diameter and full skin thickness (approximately 10 mm). RNA samples from 12 animals (3 tick-naive Holstein-Friesian, 3 tick-naive Brahman, 3 tick-infested Holstein-Friesian and 3 tick-infested Brahman) were processed and hybridised to individual slides.

Characterisation of reproductive tract microbiome and immune biomarkers for Bovine Genital Campylobacteriosis in vaccinated and unvaccinated heifers

Project description:Bovine genital campylobacteriosis (BGC) is a globally important venereal disease of cattle caused by Campylobacter fetus subspecies (C. fetus) venerealis. Diagnosis of BGC is highly challenging due to the lack of accurate diagnostic tests. To characterise the immune biomarkers for C. fetus venerealis infection, a total of twelve six cycling heifers were selected and categorised as vaccinated (n=6) with Vibrovax® (Zoetis™) and unvaccinated (n=6). All heifers were oestrous synchronised with a double dose of prostaglandin (PGF2) 11 days apart and when in oestrous intravaginally challenged with 2.7x109 CFU live C. fetus venerealis. Relative abundances of serum proteins were calculated using sequential window acquisition of all theoretical fragment ion spectra coupled to tandem mass spectrometry (SWATH-MS) for all heifers at three timepoints: pre-challenge, post-challenge and post-recovery.

R. microplus expression profiles of larvae pre-attachment and feeding adult female stages in B. indicus and B. taurus

Project description:Comparative microarray analysis of Rhipicephalus (Boophilus) microplus expression profiles of larvae pre-attachment and feeding adult female stages on Bos indicus and B. taurus cattle Global analysis of gene expression changes in R. microplus during larval, pre-attachment and early adult stages of its life cycle feeding on Bos indicus and Bos taurus cattle were compared using gene expression microarray analysis. Among the 13 601 R. microplus transcripts from BmiGI Version 2 we identified 297 up and 17 down regulated transcripts were differentially expressed between R. microplus feeding on tick resistant cattle [Bos indicus (Brahman)] compared to R. microplus feeding on tick susceptible cattle [Bos taurus (Holstein-Friesian)]. These include genes encoding enzymes involved in primary metabolism, and genes related to stress, defence, cell wall modification, cellular signaling, receptor and cuticle. Microarrays were validated by qRT-PCR analysis of selected transcripts including the validation of three housekeeping genes. The analysis of all tick stages under survey suggested a coordinated regulation of defence proteins, proteases, and protease inhibitors to achieve successful attachment and survival of R. microplus on different host breeds particularly Bos indicus cattle. The microarray was conducted by NimbleGen Systems Inc following the method reported by Saldivar [Saldivar L et al., Insect Mol Biol 2008, 17(6):597-606]. 10 samples: 2 larva, 2 pre-attachment larva in B. indicus and 2 in B. taurus, and 2 adult ticks in B. indicus and 2 in B. taurus
