Project description:Genetic male sterility (GMS) in cotton (Gossypium hirsutum) plays an important role in the utilization of hybrid vigor. However, the molecular mechanism of the GMS is still unclear. While numerous studies have demonstrated that microRNAs (miRNA) regulate flower and anther development, whether different small RNA regulations exist in GMS and its wild type is unclear. To investigate the global expression and complexity of small RNAs during cotton anther development, three small RNA libraries were constructed from the anthers of three development stages each from fertile wild type (WT) and its GMS mutant cotton. Examination of different miRNA profiles in 2 lines.

Transcription profiling of Arabidopsis loss of function mutant AtLEJ1

Project description:The loss-of-function mutant of AtLEJ1 had well-developed anther cells and showed an increased accumulation of JA and ET in its flower bud clusters. Conversely, overexpression of AtLEJ1 caused a delay in the development of the floral organs, arrested anther cell development and secondary cell-wall biosynthesis in the endothecium cells, and reduced JA and ET contents in the flower bud clusters, resulting in male-sterility through anther indehiscence. We used microarrays to gain a comprehensive understanding of the transcriptional changes induced by AtLEJ1 gene expression, Experiment Overall Design: We performed a full genome microarray analysis (ATH1 Affymetrix chips) with the wild type, 35S:AtLEJ1/5-8 line and atlej1 mutant. Total RNAs were obtained separately from unopened flower buds at stages 1~12, when the anthers are not yet opened, and from open flowers at stages 13~14, when anthesis occurs.

micro-proteomics experiment for single meiotic-phase barley anthers

Project description:We describe here a new micro-proteomics workflow for the analysis of individual barley anthers. The workflow detects more than 4000 proteins from small amounts of material representing single or paired anther samples, covering a dynamic range of protein expression levels across five orders of magnitude. We consistently identified ~2800 proteins in a single 0.6mm anther and ~4000 proteins in a paired-anther sample. This demonstrates an important technological advance in plant proteomics, considering the limited amount of starting material. We employed our micro-proteomics workflow to investigate the proteome of the developing barley anther containing male gametes in the early stages of meiosis. We successfully identified several low abundant meiosis-related proteins, proving that our approach is highly sensitive and consequently powerful. To date, no work has been published previously on precisely staged individual plant anthers.

Project description:Protein lysine acetylation (KAC) is a dynamic and reversible post-translational modification, playing important biological roles in many organisms.Here, we reported results from a proteomic investigation to detect KAC status of the developing rice anthers near the time of meiosis (RAM), providing strong biochemical evidence for roles of many KAC-affected proteins during rice anther development and meiosis. We identified a total of 1,354 KAC sites in 676 proteins.
