Discovery of novel proteins in Agrobacterium tumefaciens to improve molecular pharming yields of Trastuzumab

ABSTRACT: PlantForm Inc. (https://www.plantformcorp.com) is a Canadian biopharmaceutical company that produces antibody drugs using molecular pharming, where a genetically modified strain of Agrobacterium tumefaciens (strain At564A, derived from C58) transforms a genetically modified strain of Nicotiana benthamiana KDFX plant line (derived from USDA accession “TW16”) for biopharmaceutical expression. However, further research is required to comprehensively profile the proteome and secretome of At564A in their pipeline to identify areas of improvement. Using mass spectrometry-based bottom-up proteomics, we identified significantly altered abundance of virulence proteins after inducing virulence using acetosyringone and plant leaf extract from Nicotiana benthamiana.

Project description:Six different Solanaceae species, Potato (Solanum tuberosum), Tomato (Lycopersicum esculentum), Pepper (Capsicum annuum), Tobacco (Nicotiana tabacum), Petunia and Nicotiana benthamiana were grown at 25C, 16h light and 8h darkness. Analysis of Colletotrichum orbiculare infection of N. benthamiana

Project description:Species from the genus Colletotrichum are the causal agents of anthracnose which contribute to significant losses to the production of commercially grown crops. The genomes of Colletotrichum orbiculare, which infects cucurbits and Nicotiana benthamiana, as well as Colletotrichum gloeosporioides, which infects a wide range of fruits and vegetables, were sequenced. A custom microarray was designed for Colletotrichum orbiculare and used to assess gene expression during infection of Nicotiana benthamiana. Gene expression of Colletotrichum orbiculare growing on its host Nicotiana benthamiana was assessed at 24 hours post inoculation, 3 days post inoculation and 7 days post inoculation. Mycelia growing in vitro and ungerminated conidia were used as controls. Three replicates were performed for each time point.

Discovery of Mouse Spleen Signaling Responses to Anthrax using Label-Free Quantitative Phosphoproteomics via Mass Spectrometry

Project description:From article abstract: Inhalational anthrax is caused by spores of the bacterium Bacillus anthracis (B. anthracis), and is an extremely dangerous disease that can kill unvaccinated victims within 2 weeks. Modern antibiotic-based therapy can increase the survival rate to ~50%, but only if administered presymptomatically (within 24-48 h of exposure). To discover host signaling responses to presymptomatic anthrax, label-free quantitative phosphoproteomics via liquid chromatography coupled to mass spectrometry was used to compare spleens from uninfected and spore-challenged mice over a 72 h time-course. Late-stage (48-72 h post-challenge) Sterne strain (lethal) infections resulted in global alterations to the spleen phosphoproteome. In contrast, delta-Sterne strain (asymptomatic; missing the anthrax toxin) infections resulted in 188 (5.8%) significantly altered (q<0.05) phosphopeptides. Twenty-six highly tentative phosphorylation responses to early-stage (24 h post-challenge) anthrax were discovered (q<0.5), and ten of these originated from eight proteins that have known roles in the host immune response. These tentative early-anthrax host response signaling events within mouse spleens may translate into presymptomatic diagnostic biomarkers of human anthrax detectable within circulating immune cells, and could aid in the identification of pathogenic mechanisms and therapeutic targets.
