CHEMICAL PROTEOMIC ANALYSIS OF ENDOCANNABINOID HYDROLASE ACTIVITY IN NIEMANN-PICK TYPE C MOUSE BRAIN

Project description:The endocannabinoid system is considered to be an endogenous protective system in various neurodegenerative diseases. Niemann-Pick Type C is a neurodegenerative disease in which the role of the endocannabinoid system has not been studied yet. Here, we report the endocannabinoid hydrolase activity in brain proteomes of a Niemann-Pick type C mouse model as measured by activity-based protein profiling. Diacylglycerol lipase α, α/β-hydrolase domain-containing protein 4 (ABHD4), ABHD6, ABHD12, fatty acid amide hydrolase and monoacylglycerol lipase activities were quantified. Chemical proteomics showed no difference in endocannabinoid hydrolase activity in the brain of wildtype compared to Niemann-Pick C1 protein (NPC1) knockout mice. Three lysosomal serine hydrolases were identified with increased activity in NPC1 knockout mouse brain: retinoid-inducible serine carboxypeptidase, cathepsin A and palmitoyl-protein thioesterase 1, and we conclude that these might be interesting therapeutic targets for future validation studies.

Xanthocillin X Effectively Kills A.Baumannii via Dysregulation of Heme Biosynthesis

Project description:Isonitrile natural products exhibit promising antibacterial activities, however, their mode of action (MoA) remains largely unknown. Based on the nanomolar potency of xanthocillin X (Xan) against diverse difficult-to-treat Gram-negative bacteria, including the critical priority pathogen Acinetobacter baumannii, we performed in-depth studies to decipher its MoA. While neither metal binding nor cellular protein targets were relevant for Xan´s antibiotic effects, sequencing of resistant strains revealed a conserved mutation in the heme biosynthesis enzyme porphobilinogen synthase (PbgS). This mutation caused impaired enzymatic efficiency indicative of reduced heme production. This discovery led to the validation of an untapped mechanism by which direct heme sequestration of Xan prevents its binding into cognate enzyme pockets resulting in uncontrolled cofactor biosynthesis, accumulation of porphyrins and corresponding stress with deleterious effects for bacterial viability. Thus, Xan represents a promising antibiotic displaying activity even against multidrug resistant strains while exhibiting low toxicity to human cells.

Repurposing human kinase inhibitors to create an antibiotic active against drug-resistant Staphylococcus aureus, persisters and biofilms

Project description:To combat methicillin-resistant Staphylococcus aureus (MRSA) infections novel drugs addressing unprecedented and resistance-free targets are desperately needed. From a screen of human kinase inhibitors, we find that the anti-cancer drug sorafenib effectively kills MRSA strains. By dissection of the sorafenib scaffold and systematic synthesis of 72 analogs, we identify a potent compound – PK150 – exhibiting a minimal inhibitory concentration of 300 nM against several MRSA strains. The antibiotic induced rapid killing of S. aureus, including challenging persisters, and eradicated established biofilms. PK150 holds promising therapeutic potential as it did not induce in vitro resistance and exhibited good oral bioavailability and in vivo efficacy in a mouse infection model. Mode of action analysis by chemical proteomics revealed several targets including stimulation of protein secretion by signal peptidase IB (SpsB). Enhanced levels of extracellular proteins and resulting imbalances in secreted autolysin levels of PK150-treated bacteria support this target hypothesis. The associated antibiotic effects, especially the lack of resistance development, likely stem from the polypharmacology attribute of the compound. PK150 is therefore the founding member of a new class of highly active antibiotics.

Neocarzilin A is a potent inhibitor of cancer cell motility targeting VAT-1 controlled pathways

Project description:The natural product neocarzilin A (NCA) was discovered decades ago and despite its potent cytotoxic effects no mode of action studies were performed up to date. Synthesis of neocarzilins A, B, C and a stereoisomer of NCA provided insights into structural preferences as well as access to probes for functional studies. NCA pertained as the most active member and was not only effective against cell proliferation but also migration, a novel and so far overlooked activity. The potent anti-migratory effects could also be confirmed in an in vivo breast carcinoma mouse model. To decipher the molecular mode of action, we applied chemical proteomics for target discovery and revealed that NCA interrogates cancer cell migration via irreversible binding to the largely uncharacterized synaptic vesicle membrane protein VAT-1. A corresponding knockout of the protein confirmed the phenotype and pull-down studies showed the interaction with an intricate network of key migration mediators such as Talin-1. Overall, we introduce VAT-1 as a promising novel target for the development of selective migration inhibitors with the perspective to limit toxicity in absence of anti-proliferative effects.

Ethynyl-1,2,3-triazolyl-phosphinate based electrophiles for cysteine labelling

Project description:Herein we report the density functional theory (DFT) guided discovery of ethynyl-triazolyl-phosphinates (PT) as a new class of electrophilic warhead for cysteine selective bioconjugation. Using DFT-calculations, we found that both the electron withdrawing character as well as the π-acidity of the P-bound triazole significantly contribute to its enhanced reactivity towards thiols. We developed a straightforward synthetic route starting from readily available organic azides and diethynyl phosphinates using CuI-catalysed azide alkyne cycloaddition in aqueous buffer to access a variety of functional electrophiles. These reagents were used to obtain fluorescent peptide-conjugates for receptor labelling on live cells and in the generation of stable and biologically active antibody-drug-conjugates. Moreover, we were able to incorporate PT-electrophiles into azide-containing proteins under native conditions and demonstrate their potential in protein-protein conjugation. Finally, we showcase the excellent cysteine selectivity of this new class of electrophile in mass spectrometry based, proteome-wide cysteine profiling.
