Proteomics analysis in HL-1 cells

ABSTRACT: DUBs are essential for maintaining protein stability and functionality, playing significant roles in myocardial I/R injury. In this regard, an analysis of proteomics analysis was performed to identify the potential substrates of MYSM1 in cardiomyocytes transfected with Flag-EV or Flag-MYSM1 plasmids.

Screening of host proteins interacting with African swine fever virus MGF360-9L

Project description:To identify the potential host protein binding to African swine fever virus MGF360-9L, the 3 × FLAG tag MGF360-9L plasmid and empty FLAG were transfected into PK-15 cells for co-immunoprecipitation and LC-MS analysis. The cell lysates were immunoprecipitated using anti-FLAG antibody. Immunoprecipitation samples of cells transfected with empty FLAG were used as negative controls to eliminate non-specific interactions. LC-MS identified the interaction between MGF360-9L and PK-15 cell proteome. The final MGF360-9L binding protein data eliminated the nonspecific interaction through empty FLAG control. The interactions that remained were analyzed by significance analysis of interactome. Finally, 268 cellular proteins interacting with MGF360-9L were identified.

PPP1R2 (Inhibitor 2) stimulates protein phosphatase PP1 through stabilisation of dynamic subunit interactions

Project description:Protein Ser/Thr phosphatase PP1 is always associated with one or two regulatory subunits or RIPPOs. One of the earliest evolved RIPPOs is PPP1R2, also known as Inhibitor-2. Since its discovery nearly 5 decades ago, PPP1R2 has been variously described as an inhibitor, activator or (metal) chaperone of PP1, but it is still unknown how PPP1R2 affects the function of PP1 in intact cells. Here, using newly developed research tools, we demonstrate that PPP1R2 stabilizes a subgroup of PP1 holoenzymes, exemplified by PP1:RepoMan, thereby promoting the dephosphorylation of their substrates. Mechanistically, the recruitment of PPP1R2 disrupts the inhibitory, fuzzy interaction between the C-terminal tail and catalytic domain of PP1, and generates an additional C-terminal RepoMan-interaction site. The resulting holoenzyme is further stabilized by a direct PPP1R2:RepoMan interaction, which renders it refractory to competitive disruption by RIPPOs that do not interact with PPP1R2. Our data demonstrate that PPP1R2 modulates the function of PP1 by altering the balance between holoenzymes through stabilisation of specific subunit interactions.

microRNA binding by wildtype Ago2 and mutant Ago2 in human cells

Project description:Ago2 binds mature microRNAs (miRNAs) to regulate gene expression. A mutant Ago2 is constructed to render it unable to bind miRNAs and inhibit mRNA translation. Human 293T cells were transiently transfected with a plasmid that encoded a FLAG-tagged wildtype human Ago2 or mutant Ago2. FLAG IP was preformed, and assoicated RNA was isolated and subjected miRNA microarray analysis.

Transformation of elongation factor 1Bdelta into heat shock response transcription factor by alternative splicing

Project description:Protein translation factors play crucial roles in a variety of stress responses. Here, we show that the eukaryotic elongation factor 1Bdelta (eEF1Bdelta) changes its structure and function from a translation factor into a heat shock response transcription factor by alternative splicing. While eEF1Bdelta is specifically localized in the cytoplasm, the long isoform of eEF1Bdelta (eEF1BdeltaL) is localized in the nucleus and induces heat shock element (HSE)-containing genes in cooperation with heat-shock transcription factor 1 (HSF1). Moreover, the N-terminal domain of eEF1BdeltaL binds with NF-E2-related factor 2 (Nrf2) and induces stress response heme oxygenase 1 (HO-1). Specific inhibition of eEF1BdeltaL with siRNA completely inhibits Nrf2-dependent HO-1 induction. In addition, eEF1BdeltaL directly binds to HSE oligo DNA in vitro and associates with HSE containing the HO-1-enhancer region in vivo. Thus, the transcriptional role of eEF1BdeltaL could provide new insights into the molecular mechanism of stress responses. Project description:High-grade serous ovarian cancer (HGSOC) is the deadliest gynecologic malignancy in women.The lack of effective second line therapeutics remains a substantial challenge for BRCA-1/2 wildtype HGSOC patients, and contributes to poor survival rates due to drug resistance. There is a striking need to elucidate and implement new and alternative treatment options for patients with HGSOC. Histone Deacetylases (HDACs) are promising targets in HGSOC treat-ment, however, the mechanism and efficacy of HDAC inhibitors is understudied in HGSOC. In order to consider HDACs as a treatment target, we need to better understand how they are functioning within HGSOC. This includes elucidating HDAC6 protein-protein interactions. Mass spectrometry runs and analysis of host proteins co-immunoprecipitated with RABV M protein

Project description:To explore the role of the matrix protein in RABV infection of host cells, we looked for host proteins that interact with the RABV M protein. We performed Flag-tag-based immunoprecipitation of lysates of HEK293T cells transfected with plasmids expressing either N- or C-terminally Flag-tagged M protein (M-NF or M-CF, respectively), and identified host proteins that interacted with the M proteins by using mass spectrometry (MS). Proteins were detected in lysates of cells transfected with the plasmid expressing Flag-tagged M proteins but not in those of cells transfected with the control plasmid. We identified a total of 2,672 host proteins that co-immunoprecipitated with the M protein; 492 proteins co-immunoprecipitated to both M-NF and M-CF, 1,761 proteins co-immunoprecipitated with M-NF only, and 419 proteins co-immunoprecipitated with M-CF only.

mRNA 3â€™UTR lengthening by alternative polyadenylation attenuates inflammatory responses and correlates with virulence of Influenza A virus

Project description:Alternative polyadenylation (APA) leading to changes of mRNA 3â€™UTRs has been associated to numerous pathologies but its molecular origin and functional consequences often remain enigmatic. We show that all Influenza A virus (IAV) strains cause APA of host transcripts and mapped the effect to a drug-targetable glycine residue (G184) in the viral non-structural protein 1 (NS1) that is required for its interaction with CPSF4. Unexpectedly, this interaction is also conserved in IAVs not causing virus-induced transcriptional shutoff and introduction of G184R in NS1 alleviates APA. Functionally, a recombinant PR8 (NS1-G184R) virus elicited increased secretion of inflammatory cytokines as compared to the parental wild-type in vitro and in vivo, and had a strongly attenuated phenotype in vivo. Finally, we show that many pathogens induce APA, which is potentially used towards similar immunomodulatory effects.
