Project description:Some insects can redirect plant development to form unique organs called galls, which provide these insects with unique, enhanced food and protection from enemies and the elements. Many galls resemble flowers or fruits, suggesting that elements of reproductive development may be involved. We addressed this possibility using RNA sequencing (RNAseq) to quantify the transcriptional responses of wild grapevine (Vitis riparia Michx.) leaves to a galling parasite, phylloxera (Daktulosphaira vitifoliae (Fitch 1855)). If development of reproductive structures is part of gall formation, we expected to find significantly elevated expression of genes involved in flower and/or fruit development in developing galls as opposed to ungalled leaves. We found that reproductive gene ontology classes were significantly enriched in developing galls, and that expression of many putative genes involved in flower formation was significantly increased, particularly in later gall stages. The patterns of gene expression found in galls suggest that phylloxera exploits vascular cambium to provide meristematic tissue and redirects leaf development towards formation of carpels. The phylloxera leaf gall, and perhaps other similar galls, appears to be phenotypically and transcriptionally convergent on the plant carpel.

Project description:Oak galls form when gall wasps lay their eggs into part of the tree; in some galls, this attachment point to the host consists of only a few cells. The gall itself comprises entirely of host tissue; however, the initiation, development, and physical appearance are controlled by the inducer. This raises the intriguing question of the molecular mechanisms underlying gall formation, by which one or a small number of cells are reprogrammed and commit to a novel developmental path. Gall wasps undergo two generations each year, and the galls formed by these two generations exhibit markedly different appearances. We sequenced the transcriptomes of both the sexual and asexual generations of Neuropterus quercusbaccarum and Neuroterus numismalis. The transcriptomes of the generations that occur at the same time of year are more similar to each other than they are to the opposite generation of their respective species.

Project description:Understanding how plant tissue and organs may be transformed into novel structures by other organisms provides a unique opportunity to study the molecular processes that dictate facets of plant anatomy and morphology. Certain groups of wasps have evolved the ability to transform plant tissues into ornate structures called galls, which provide shelter and nutrition for their larvae. However, the exact mechanism for how gall wasps remodel the plant’s physical structure and metabolism is still largely unknown. At their core, galls alter the morphology and repurpose the function of plant tissue. One common trait that unites all galls is the distinct cellular reprogramming and tumor-like growth that is necessary to produce a gall. There are over 1,400 gall wasps from the family Cynipidae, resulting in a wide diversity of gall structures, shapes, and colors that have been described. Thus, discovering the core molecular determinants that dictate the radical transformation of plant cells will help reveal principles of how plant morphology and function can be rewired by external factors. Little molecular work has been done to elucidate the factors (i.e., genes, proteins, or small molecules) that may be involved in this dramatic repurposing and dedifferentiation of plant tissue. We plan to utilize modern -omics approaches to investigate and identify the molecular factors that underlie the initiation and development of plant galls.

Project description:BackgroundThe tree of life is significantly asymmetrical - a result of differential speciation and extinction - but general causes of such asymmetry are unclear. Differences in niche partitioning are thought to be one possible general explanation. Ecological specialization might lead to increases in diversification rate or, alternatively, specialization might limit the evolutionary potential of specialist lineages and increase their extinction risk. Here we compare the diversification rates of gall-inducing and non-galling insect lineages. Compared with other insect herbivores feeding on the same host plant, gall-inducing insects feed on plant tissue that is more nutritious and less defended, and they do so in a favorable microhabitat that may also provide some protection from natural enemies. We use sister-taxon comparisons to test whether gall-inducing lineages are more host-specific than non-galling lineages, and more or less diverse than non-gallers. We evaluate the significance of diversity bipartitions under Equal Rates Markov models, and use maximum likelihood model-fitting to test for shifts in diversification rates.ResultsWe find that, although gall-inducing insect groups are more host-specific than their non-galling relatives, there is no general significant increase in diversification rate in gallers. However, gallers are found at both extremes - two gall-inducing lineages are exceptionally diverse (Euurina sawflies on Salicaceae and Apiomorpha scale insects on Eucalytpus), and one gall-inducing lineage is exceptionally species-poor (Maskellia armored scales on Eucalyptus).ConclusionsThe effect of ecological specialization on diversification rates is complex in the case of gall-inducing insects, but host range may be an important factor. When a gall-inducing lineage has a host range approximate to that of its non-galling sister, the gallers are more diverse. When the non-galler clade has a much wider host range than the galler, the non-galler is also much more diverse. There are also lineage-specific effects, with gallers on the same host group exhibiting very different diversities. No single general model explains the observed pattern.

Project description:Priming of plant defenses provides increased plant protection against herbivores and reduces the allocation costs of defense. Defense priming in woody plants remains obscure, in particular due to plant development traits such as the endogenous rhythmic growth displayed by oaks (Quercus robur). By using bioassays with oak microcuttings, and by combining transcriptomic and metabolomic analyses, we investigated how leaf herbivory by Lymantria dispar and root inoculation with the ectomycorrhizal fungus Piloderma croceum prime oak defenses. We further investigated how defense priming is modulated by rhythmic growth of the oaks. A first herbivory challenge in oak leaves primed newly grown leaves for an enhanced induction of jamonic acid (JA)-related direct defenses, or enhanced emission of volatiles, depending on the specific growth stage at which the plants where challenged. Root inoculation with Piloderma abolished the enhanced induction of JA-related defenses and volatile emission. Our results indicate that a first herbivore attack primes direct and indirect defenses of newly formed oak leaves, and that the specific display of defense priming is modulated by rhythmic growth. Our results further show that the priming memory in oaks can be transmitted to the next growth cycle even to the leaves of the new shoot unit.

Project description:Pantoea agglomerans pv. betae (Pab) induces tumor-like galls in beet and gypsophila, a process mediated by the secretion of effector proteins via Pab's type III secretion system (T3SS). However, the molecular mechanisms underlying Pab-induced gall formation remain largely unexplored. This study delves into the cellular architecture and transcriptional profile of Pab-mediated galls, comparing host responses to wild-type Pab and a T3SS-inactive mutant, hrcC-. Morphological analysis using scanning electron microscopy and cross-sectional visualization of infected beet leaf tissues revealed that Pab-induced gall-like structures are linked to cell hyperplasia and tissue ruptures, contingent on T3SS activity. Comparative transcriptome analysis of wild-type Pab and hrcC- Pab-infected beet leaves at 12 and 48 hours unveiled significant transcriptional reprogramming, with nearly 2,000 differentially expressed genes as of 48 hours post inoculation. Enrichment analyses identified the upregulation of pathways related to signal transduction, defense, carbohydrate metabolism, and cell wall modulation in wild-type Pab-infected leaves compared to controls. Particularly notable was the significant upregulation of numerous genes associated with cell wall loosening by wild-type Pab, suggesting an initial rearrangement of cell wall architecture facilitating gall formation. Furthermore, transcriptome analysis demonstrated that wild-type Pab suppresses the expression of the betalain biosynthetic gene DOPA 4,5-DIOXYGENASE, leading to reduced betalain accumulation in infected tissues compared to the mutant strain. These findings offer fresh insights into the transcriptional and physiological manipulation of host tissue during the early stages of Pab-induced gall formation.
